摘  要:目的观察罗格列酮对糖基化终产物(AGEs)干预的人肾小球系膜细胞(HRMC)β1-整合素表达以及细胞外基质(ECM)成分Ⅳ型胶原分泌的影响。方法用葡萄糖和牛血清白蛋白制备糖基化终产物(AGE-BSA),用不同浓度的罗格列酮(1.0、10.0、100.0μmol/L)和AGE-BSA共同干预以及AGE-BSA单独干预(AGEs组)HRMC。RT-PCR法测定细胞β1-整合素mRNA水平;Western blot法检测β1-整合素蛋白表达;放射免疫分析法测定细胞培养上清液Ⅳ型胶原含量。结果与AGEs组相比,各浓度罗格列酮均能下调由AGE-BSA诱导的HRMCβ1-整合素mRNA和蛋白表达的升高,并抑制培养上清液Ⅳ型胶原的分泌(P<0.05)。结论罗格列酮可能通过抑制AGEs引起的HRMCβ1-整合素mRNA和蛋白表达及Ⅳ型胶原含量的升高,发挥一定的肾脏保护作用。Objective:To observe the influence of rosiglitazone on the expression of β1-integrin and the secretion of type Ⅳ collagen in cultured human renal mesangial cells(HRMCs) incubated with advanced glycation end products(AGEs).Methods:HRMCs were incubated with AGEs modified bovine serum albumin(AGE-BSA),which were treated with different concentrations(1.0,10.0,100.0 μmol/L) of rosiglitazone.The mRNA expression of β1-integrin in HRMCs was analyzed by RT-PCR,and the protein expression of β1-integrin in HRMCs was determined by Western blot.The content of type Ⅳ collagen in cell culture media was measured by radioimmunoassay.Results:Rosiglitazone decreased the levels of β1-integrin mRNA and protein in HRMCs and the level of type Ⅳ collagen in supernatants of cultured HRMCs incubated with AGE-BSA(P0.05).Conclusion:Rosiglitazone may have renoprotective effect through inhibiting the increases of β1-integrin expression and Ⅳ collagen cotent.
