摘 要:目的观察人脐血间充质干细胞(MSCs)对大鼠脊髓损伤的治疗作用,为脊髓损伤的临床治疗提供新策略。方法分离纯化人脐血MSCs,制备大鼠脊髓损伤模型,随机分为假手术组、模型组和MSCs移植组(BrdU标记),移植后7、14、21d,采用BBB评分评估大鼠行为学变化,免疫荧光法检测MSCs的迁移、存活情况,免疫组化法检测炎症因子HMGB1、NF-κB及凋亡蛋白Caspase-3在脊髓损伤部位的表达变化。结果移植后21d,MSCs移植组大鼠肢体功能恢复明显,与模型组比较差别有统计学意义(P<0.05)。MSCs移植组大鼠脊髓损伤区及周边可见BrdU阳性细胞。相同时间点,MSCs移植组HMGB1、NF-κB和Caspase-3的阳性表达率远低于模型组,差别有统计学意义(P<0.05)。结论人脐血MSCs移植可显著促进大鼠脊髓损伤的康复,这可能为脊髓损伤的临床治疗提供一种新的途径。Objective To observe the therapeutic effect of mesenchymal stem cells(MSCs) derived from human umbilical cord blood to treat rat spinal cord injury and to provide new tactics for clinical therapy.Methods MSCs were separated and purified from human umbilical cord blood.The spinal cord semi-traverse models were established and randomly divided into three groups:sham-operation group,model group and MSCs transplantation group(BrdU labeled).Functional outcome measurements using the Basso-Beattie-Bresnehan(BBB) score were performed periodically upto post-transplantation 21d.MSCs survival and migration were observed by immunofluorescence techniques.The expression of HMGB1,NF-κB and caspase-3 were detected by immunohistochemical techniques at various time points after transplantation.Results BBB scores showed that locomotor recovery improvement in MSCs transplantation groups was greater than that in model group post-transplantation 21d(P0.05).BrdU-labeled MSCs could be seen in spinal cord injury area.The positive rate of HMGB1,NF-κB and caspase-3 expression in MSCs transplantation group were lower than that in model group,and there was a significant difference between them(P0.05).Conclusion The transplantation of MSCs derived from human umbilical cord blood could promote functional recovery of the injuried spinal cord dramatically,which provide a new way for clinical therapy.
