摘  要:目的:观察大蒜素对糖尿病大鼠心肌细胞凋亡及MAPK信号传导通路的影响。方法:应用链脲佐菌素建立糖尿病大鼠模型,随机分为糖尿病模型组、大蒜素低、中和高剂量组,同时设正常对照组。大蒜素各剂量组按不同浓度比例喂饲大蒜素。连续干预16天后处死剪取左心室组织,TUNEL检测心肌细胞凋亡情况,Western blotting检测MEK、ERK和P38表达情况。结果:模型组心肌细胞凋亡程度较重,大剂量组心肌细胞凋亡情况改善明显;模型组MEK、ERK表达下降,P38表达增加,大剂量组的表达改善明显。结论:大蒜素可有效防治糖尿病心肌细胞凋亡,激活MEK-ERK通路以保护心肌,同时降低P38蛋白表达。Objectives: To observe the effect of allicin on apoptosis and MAPK of heart cell in diabetic rats.Methods: Diabetes models were created by injecting rats with Streptozocin,all the model rats were randomly divided into model group,high-dose group,middle-dose group and low-dose group,and fed with different dose of allicin,meanwhile control group was observed.After 16 days of feeding,all the rats were killed and samples were gathered to observe apoptosis of heart cell and the expression of MEK,ERK and P38.Results: Severe apoptosis,lower expression of MEK and ERK,and higher expression of P38 could be observed in model group.In high-dose group,such changes were improved.Conclusion: Allicin can effetely prevent apoptosis,protect heart cell by activating expression of MEK-ERK and lowering expression of P38.
