摘 要:考察麦芽糖诱导木聚糖酶基因在枯草芽孢杆菌WB700中的表达。以麦芽糖为诱导剂,对诱导质量分数、诱导时机、诱导温度和麦芽糖的添加方式等主要因素进行了分析比较。结果表明,当菌体生长至发酵液吸光值OD600达到1.3时加入终质量分数为4.5%麦芽糖,37℃,持续诱导10 h,木聚糖酶活力达到最高,为56.32 U。通过SDS-PAGE检测对比显示:经麦芽糖诱导表达的木聚糖酶比不加麦芽糖诱导的木聚糖酶表达量高,表达效果更明显。Attempt to use maltose to induce the expression of alkali-tolerant xylanase by Bacillus subtilis WB700,the main factors,such as the concentration of the inducer,the right time and temperature of adding the inducer and the adding method,was studied.The result showed that,when the OD600 absorbance of fermentation broth reached to 1.3,adding maltose with the final concentration of 4.5%,and induced at 37 ℃for 10 h,xylanase activity of the cells harvested the maximum,56.32 U.Compared the xylanase expressed under inducing by maltose with the xylanase without inducing through SDS-PAGE,it indicated that the expression lever of the former was high than that of the latter.
