白介素18对结肠癌sw480细胞移植瘤HIF-1α表达的影响

摘　　要：目的观察人白介素18(hIL-18)对结肠癌sw480细胞的抑制效应及hIL-18对sw480细胞移植瘤缺氧诱导因子-1α(HIF-1α)表达的影响。方法噻唑蓝(MTT)法及流式细胞术(FCM)检测结肠癌sw480细胞、转染携带绿色荧光蛋白表达质粒的pEGFP-sw480细胞、转染携带hIL-18基因表达质粒(pEGFP)的hIL-18-pEGFP-sw480细胞,观察hIL-18对细胞增殖及细胞周期的影响。建立sw480、pEGFP-sw480、hIL-18-pEGFP-sw480 3组细胞株裸鼠移植瘤模型。免疫组织化学法检测移植瘤组织中HIF-1α的表达情况。结果根据MTT法绘制的细胞生长曲线示hIL-18-pEGFP-sw480组较pEGFP-sw480组、sw480组到达平台期延长,hIL-18明显抑制了细胞的增殖速率(P<0.05)。FCM术结果显示hIL-18-pEGFP-sw480组细胞G0/G1期比例显著高于pEGFP-sw480组、sw480组,S、G2/M期比例明显低于pEGFP-sw480组、sw480组(P<0.05)。hIL-18-pEGFP-sw480组HIF-1α灰度值高于SW480、pEGFP-sw480组,差异有统计学意义(P<0.05)。结论 IL-18能够抑制结肠肿瘤HIF-1α的表达。Objective To investigate the effects of the recombinant eukaryotic plasmid hIL-18-pEGFP exprssing human IL-18 gene to sw480 cells and the effects of hIL-18 to the expression of hypoxia inducible factor 1α（HIF-1α） in xenografts tumor.Methods Investigate inhibitive effect of hIL-18 to the growth of sw480 cells by MTT,and impact on cell cycle of sw480 cells by FCM.To establish nude mouse tumor models of sw480、pEGFP-sw480 and hIL-18-pEGFP-sw480 cells and HIF-α protein of tumor xenografts was assayed by immunocytochemical technique.Results The growth curve showed that the growth of hIL-18-pEGFP-sw480 cells were obviously inhibited,there was a statistical difference compare with sw480 cells and pEGFP-sw480 cells（P0.05）.The result of FCM showed the percerntage of G0/G1 phase of hIL-18-pEGFP-sw480 cells was higher than sw480 cells and pEGFP-sw480 cells（P0.05）,the percerntage of S.G2/M phase of hIL-18-pEGFP-sw480 cells was lower than sw480 cells and pEGFP-sw480 cells（P0.05）.Immunocytochemical technique indicated that the gray value of hIL-18-pEGFP-sw480 group about HIF-1α was more than sw480、pEGFP-sw480 groups（P0.05）.Conclusion IL-18 could inhibit the expression of HIF-1α.in colon carcinoma
