P53蛋白和增殖细胞核抗原在涎腺良恶性肌上皮瘤中的表达

摘　　要：目的：检测 Ｐ５３ 蛋白和 Ｐ Ｃ Ｎ Ａ 在涎腺良恶性肌上皮瘤中的表达。方法：采用免疫组化方法检测１２ 例肌上皮瘤、８ 例肌上皮瘤细胞生长活跃型、２０ 例恶性肌上皮瘤中 Ｐ５３ 蛋白和 Ｐ Ｃ Ｎ Ａ的表达，用图像分析系统检测 Ｐ Ｃ Ｎ Ａ 的相对含量。结果： Ｐ５３ 蛋白在６ 例恶性肌上皮瘤中呈阳性表达，在良性肌上皮瘤中均呈阴性表达； Ｐ Ｃ Ｎ Ａ阳性表达的增殖指数和相对含量在恶性肌上皮瘤中表达高于在良性肌上皮中的表达（ Ｐ＜０．０１）； Ｐ Ｃ Ｎ Ａ 高表达与 Ｐ５３ 蛋白高表达有一定相关性。结论：ｐ５３ 基因突变对恶性肌上皮瘤的发生有一定作用； Ｐ Ｃ Ｎ Ａ 阳性表达和相对含量的检测是诊断恶性肌上皮瘤的重要生物学指标。Objectlve: To study the expression of P53 and PCNA in myoepithelioma and malignant myoepithelioma of salivary gland.Methods:12 cases of myoepithelioma,8 of myoepithelioma with active proliferation and 20 of malignant myoepithelioma were examined by immunohistochemical methods.Results:P53 protein expression was positive in six of malignant myoepithelioma and negative in all cases of myoepithelioma.The proliferating index (PI) of PCNA positive cells in malignant myoepithelioma showed a higher index of nuclear staining of P53 protein (mean 22.08 %) than that in myoepithelioma(mean 4.17%) or myoepithelioma with active proliferation(mean 6.00%).The absorbance of PCNA relative content in malignant myoepithelioma ( 0.17 ±0.158) was higher than that in benign myoepithelioma (0.03±0.36 for myoepithelioma and 0.08±0.087 for that with active proliferation) ( P <0.01).Conciuslon:PCNA and the mutation of p 53 gene may play an important role in the carcinogenesis of malignant myoepithelioma.
