摘 要:目的:观察阿米替林(AMI)对大鼠皮层神经元钠通道电流及其mRNA表达的影响。方法:运用全细胞膜片钳模式记录AMI加药前后的钠通道电流(INa);采用实时逆转录聚合酶链反应(real time reverse-transcription-polymerase chain reaction,RT-PCR)观察实验组和对照组皮层神经元钠通道各亚型的mRNA表达量。结果:AMI抑制大鼠皮层神经元INa及其mRNA表达(P<0.05)。10μMAMI显著改变钠通道的激活特性和失活特性。10μMAMI在5 min、10 min及24 h对Nav1.1均有抑制作用,而对于Nav1.2的作用只是在5 min时有抑制作用,对Nav1.6的抑制作用存在波动性的变化。结论:AMI对大鼠皮层神经元INa及其各亚型mRNA的表达均有浓度依赖性抑制作用。AMI对INa的抑制主要通过影响钠通道的激活和失活动力学来实现的,对其各亚型mRNA表达的抑制作用具有不同的特点。Objective: To investigate the effect of amifriptyline(AMI) on INa and the expression of mRNA of sodium channels in cultured rat cortical neurons.Method: The effects of AMI on INa were observed by the whole-cell patch clamp recoding and the mRNA expression of different sodium channels were assessed in experimental and control groups by RT-PCR.Results: AMI significantly inhibited INa and the mRNA expression in rat cortical neurons.AMI of 10 μM significantly changed the activation and inactivation of INa.The inhibition on Nav1.1 was observed at 5 min,10 min and 24 h while Nav1.2 appeared a transient inhibition at 5 min.The inhibiton of Nav1.6 fluctuated at the different time after 10 μM AMI treatment.Conclusion: AMI has a significant inhibitory effect on INa and the expression of mRNA in rat cortical neurons in a concentration-dependent manner.The inhibition of INa was shown by affecting the activation and inactivation of the channel.AMT presented differential effects on the mRNA expression of various subtypes of sodium channels.
