摘 要:采用高速逆流色谱法分离制备粉被玛利亚霉菌丝体甲醇提取物中的天然色素。溶剂体系为V(乙酸乙酯):V(正丁醇):V(甲醇):V(水)=10:3:3:10,以其下相为流动相,上相为固定相,转速850 r/min,流速为2mL/min,紫外检测波长340 nm。所得色素产物的纯度经高效液相色谱分析为95.3%(峰面积归一化法),其分子式由高分辨ESI-MS确认为C26H32O9。经天然产物数据库查询,没有发现与该分子式匹配的化合物,因此该色素可能为一新化合物。High-speed counter-current chromatography was used for separation and purification of natural pigment from the myeelia of Mariannaea puinosa. The tow-phase solvent system was ethyl acetate-n-butanol-methanol-water (10: 3: 3: 10). The upper phase was stationary phase and the lower phase was mobile phase. The flow-rate of the mobile phase was 2.0 mL/min and the rotation speed was 850 r/min. The effluent of the column was monitored with a UV detector at 340 nm. The purity of the pigment fraction was evaluated by high-performance liquid chromatography (HPLC). High resolution MS analysis revealed that the molecular formula of the compound was C26H32O9, which was possibly a new compound according to the searching results of Chapman & Hall natural products database.
