摘  要:The in-vivo effect of ofloxacin and norfloxacin on the pharmacokinetics of isoniazid (INH) was investigated. Five apparently healthy volunteers aged 18-39 years participated in the study after getting an informed consent. The study was carried out in three phases with an interval drug wash out period of at least one week in between the phases. Phase one took 300 mg of isoniazid (usual adult dose); phase two was concurrently administered with 300 mg isoniazid plus 200 mg ofloxacin and phase three followed the same procedure with each subject receiving 300 mg isoniazid together with 400 mg norfloxacin. All medicine was taken orally with 350 ml of water. The concentrations of isoniazid (INH) in urine of the subjects at predetermined time interval were measured spectrophotometrically for the three phases over a period of 0-48 hours. The urinary excretion of isoniazid (INH) was increased by ofloxacin as well as norfloxacin; the excretion rate of INH was also increased by the fluoroquinolones and the percentage urinary excretion oflNH was 84.60 and 80.80 mg for ofloxacin and norfloxacin respectively as against 78.0 mg for the control.
