摘  要:目的观察伽玛刀照射对致痫大鼠海马组织DBP及p-ERK1/2蛋白的影响,探讨伽玛刀治疗颞叶癫痫的作用机制。方法实验分正常对照组、模型组、伽玛刀治疗组,每组30只大鼠。以匹罗卡品腹腔注射复制大鼠颞叶癫痫模型,实验设立伽玛刀治疗后3h、7d、15d、30d、60d等五个不同时间点进行指标检测,应用western blotting法检测大鼠海马组织DBP、p-ERK1/2蛋白的表达。结果匹罗卡品点燃模型后大鼠海马的DBP被显著激活,并随时间出现有规律的变化,在各时间点模型组大鼠海马区DBP蛋白的表达与正常对照组相比具有统计学差异(P<0.05),而伽玛刀放射治疗能显著抑制这种激活(P<0.05)。海马区p-ERK1/2蛋白的变化呈现与DBP蛋白相同的规律。结论伽玛刀对DBP-ERK1/2信号通路的影响可能是其治疗颞叶癫痫的作用机制之一。Objective To observe the effect of gamma knife on expression of DBP protein and p-ERK1/2 protein in hippocampal tissue in rat with epilepsy induced by pilocarpine,so that to explain the therapeutic mechanism of gamma knife on epilepsy.Methods Rats were randomly divided into 3 groups,normal control group,model group,gamma knife group,each group included 30 rats.Epileptic model was reduplicated by intraperitoneal injection with pilocarpine,protein expression of DBP and p-ERK1/2 in hippocampal tissue was determined by western blotting in 5 time points,3h,7d,15d,30d and 60d respectively.Results Pilocarpine showed significant activation of DBP with time depended manner,the expression of DBP protein was significantly increased in model group than that in normal control group(P0.05),while in gamma knife,the activation of DBP was significantly depressed(P0.05),the expression of p-ERK1/2 in hippocampal tissue was the same trend.Conclusion Activation of DBP-ERK1/2 signaling pathway maybe one of the therapeutic mechanisms of gamma knife on temporal lobe epilepsy.
