摘 要:目的探究Wnt1/β-catenin信号通路与食管鳞状细胞癌(ESCC)的临床病理及预后因素的相关性。方法用实时定量PCR(RT-PCR)和免疫组织化学技术分析70例ESCC(包括癌组织与癌旁非癌组织)中Wnt1/β-catenin的表达情况,统计分析其与临床病理及预后的关系。结果癌组织中Wnt1mRNA水平(1.9934±1.9888vs.0.8863±0.6658,P=0.0000)和蛋白质水平(0.3830±0.0947vs.0.2721±0.1474,P=0.0000)均较癌旁组织中显著高表达;Wnt1mRNA表达与病理分期和淋巴结转移及预后相关(P<0.05),但蛋白质水平未显示相关。癌组织中β-cateninmRNA水平(0.2854±0.1298vs.0.8863±0.6658,P=0.0000)和蛋白质水平(0.2835±0.0844vs.0.2352±0.0670,P=0.003)较癌旁组织中显著高表达,β-cateninmR-NA表达与病理分期、淋巴结转移及预后相关(P<0.05),但蛋白质水平并未显示相关。在蛋白质水平上,Wnt1和β-catenin表达水平无相关性。结论 Wnt1/β-catenin在ESCC组织中显著高表达,可能成为判断ESCC分期和预后的重要分子标记物。且Wnt1/β-cateninmRNA表达水平与ESCC临床病理分期、淋巴结转移呈逆相关,mRNA的异常表达者预后不佳,但可能不是独立预后因素。Objective To clarify the correlatian between Wnt 1/β-catenin expression and the clinicopathologic ehatacteristics and prognosis of patients with esophageal squamous cell carcinoma ( ESCC ). Methods Quantitative real-time polymerase chain reaction (RT-PCR) and immunohist^hemical staining for Wnt 1/±catenin were Offonned on cancerous and adjacent non-cancerous paraffin- embedded esophageal specimens. Results The level of Wnt 1 mRNA in ESCC was significantly higher than that in the adjacent non - cancerous tissues ( 1. 9934 ± 1. 9888 vs. 0. 8863±0. 665 ,P = 0. 0000 ), and its overexpression was aggressively asscwiated with lymphnode metastasis, zdvanced pathologic.,d stage and survival ( P 〈 0. 05 ). The level of Wnt I protein expression was al significantly higher than that in the adjacent non-cancerous tissues ( 0. 3830 ± 0. 0947 vs. 0. 2721 ±0. 1474, P = 0. 0002 ), however, its overexpression was not associated with lymphnode metastasis and advanced pathological stage . The level of Wnt 1 expression had no relevance with the smvival of patients. The level of β-catenin mRNA in ESCC was significantly higher than that in the adjacent non -cancerous tissues (0. 2854 ±0. 1298 vs'. 0. 8863±0. 6658, P = 0. 0000 ), and its overexpression was aggs'essively associated with lymphnode metastasis , advanced pathological stage and survival ( P 〈 0. 05 ). The level of β-catenin protein expression was significantly higher than that in the adjacent non -cancerous tissues (0. 2835 ±0. 0844 vs. 0. 2352 ± 0. 0670, P = 0. 003 ), however, its overexpression was not related to clinicopathologic characteristics . Meantime,The level of Wnt 1 protein expression had no relevance with that of β-catenin. Conclusions The overexpression of Wnt 1/β-cateniu may he the important molecular marker to predict the clinicopathologic stage and prognosis of ESCC , and the level of Wnt 1/β- catenin mRNA is inversely correlated with lymphnode metastasis and advanced pathological stage .
