The study of partial biological characteristics and identification on two plant probiotics for animals

摘  要:为了探明2株植物源芽孢杆菌K,F(暂定名)的抗逆性与抑菌特性,对其进行了耐酸,耐胆盐与抑菌试验。用pH 1.5、2.0、3.5的人工胃液模拟胃液环境;用0.3%胆盐及0.1%CuSO4.5H2O的人工肠液模拟肠液环境。分别测定K,F的培养上清与菌液抑制大肠杆菌的能力。采用传统的表型鉴定及生理生化鉴定方法及16SrDNA方法对K,F进行鉴定。结果:经过pH 1.5、2.0、3.5的胃液处理,K,F均有较高的存活率,随着pH的上升,存活率有上升的趋势;耐胆盐试验,处理组细菌存活率有所下降,但均高于90%;K的培养上清液和菌液对大肠杆菌的抑菌环直径为14.33,18.57 mm,F的为14.11,15.68 mm。所以人工胃液,人工肠液对K,F的活菌数几乎不产生影响。K,F的培养上清和菌液对大肠杆菌有很好的抑制作用。综合鉴定结果K为枯草芽孢杆菌,F为地衣芽孢杆菌。In order to study the stress resistance of two plant Bacillus K and F(temporary names),acid resistant and bile tolerance experiment had been made.Simulated stomach juice was pH 1.5,2.0 and 3.5,simulated intestinal fluid included 0.3% bile salt and 0.1% CuSO4 5H2O.The K and F's antibacterial capacity of supernatant and bacterial was liquid tested respectively.They were identified with phenotype,biochemical and 16SrDNA methods.In artifical stomach of pH 1.5,2.0,and 3.5,both K and F had high survival rates.With the increase of pH,there was a rising trend of survival rate.In the experiment of bile tolerance,there was downward trend of survival rate,but the survival rate was over 90%.The inhibition zone diameter against E.coli of K was 14.33,18.57 mm,and F was 14.11,15.68 mm.So artificial stomach juice and artificial intestinal juice had little influence on K and F's living bacterial number.The supernatant and bacterial liquid of K and F had good inhibition effect on E.coli.The comprehensive identification results were that K was Bacillus subtilis,F was Bacillus licheniformis.
