注射用银杏内酯B在比格犬体内的药代动力学研究

摘　　要：通过测定注射用银杏内酯B静脉给药比格犬血药代谢动力学，为指导临床安全、合理用药提供理论依据。对18只比格犬，静脉注射给药，给药后于不同时间在经前肢静脉采血2．5-3mL，用气相一质谱联用法（GC—MS）测定血药浓度，计算药代动力学参数。结果如下：比格犬静脉给予注射用银杏内酯B0．5，1，2mg／kg，分布半衰期t1／2α分别为4．0，2．3，1．8min，消除半衰期t1／2α分别为24．9，26．7，19．3min，药时曲线下面积AUC（0-t）分别为5．9，11．0，13．2μg／（L·min）。银杏内酯B血药浓度在0．01～4μg／mL，线性关系良好（r=0．997），犬静脉注射用银杏内酯B后，药时曲线符合二室模型。银杏内酯B呈快速分布，消除迅速，体内分布广泛。The experiment was designed to determine the pharmacokineties of injected ginkgolides B in beagle canines＇ vein in order to provide theroretical reference to guide clinical safety and rational administration of drugs. GC - MS was used to determine the blood concentration of ginkgolides B. The injected ginkgolides B in beagle canines＇vein were 0. 5,1,2mg/kg; the distribution half-time t1/22 was 4.0,2.3,1.8 min ; the elimination half - time t1/2α was 24.9,26.7,19.3 min ; and the AUC（0-t） was 5. 9,11.0,13.2μg/（ L · min）, respectively. The concentration of ginkgolides B at 0.01 - 4μg/mL had a favorable linear relationship（ r = 0.997 ）. Moreover, the disposition of ginkgolides B in blood fitted two - compartment model. Additionally, the ginkgolides B was fast -distribution, swift -elimination and wide - spread.
