摘  要:目的探讨脂多糖(LPS)干预后小胶质细胞内血小板生成素(TPO)与IL-6的表达及其相关性。方法体外培养BV2细胞,以LPS干预小胶质细胞12~24 h,实验分为6组。空白12 h组:BV2细胞正常培养12 h,不添加任何干预因素;LPS 0.5 mg.L-112 h组:在培养好的BV2细胞内添加预先配好的LPS溶液共同培养12 h,并使其终质量浓度为0.5 mg.L-1;LPS 1.0 mg.L-112 h组:在培养好的BV2细胞内添加预先配好的LPS溶液共同培养12 h,并使其终质量浓度为1.0 mg.L-1;空白24 h组:BV2细胞正常培养24 h,不添加任何干预因素;LPS 0.5 mg.L-124 h组:在培养好的BV2细胞内添加预先配好的LPS溶液共同培养24 h,并使其终质量浓度为0.5 mg.L-1;LPS 1.0 mg.L-124 h组:在培养好的BV2细胞内添加预先配好的LPS溶液共同培养24 h,并使其终质量浓度为1.0 mg.L-1。采用ELISA法检测各组细胞内IL-6及TPO水平,免疫印迹法检测细胞内IL-6及TPO蛋白表达水平。结果 LPS处理后明显增加BV2细胞内TPO及IL-6蛋白表达水平,其中0.5 mg.L-1LPS干预细胞12 h时,TPO及IL-6水平处于最高峰;1.0 mg.L-1LPS干预细胞12 h时,TPO及IL-6蛋白水平增加最明显,且二者表达呈显著正相关(r=0.876,P<0.01)。结论 TPO及IL-6参与LPS诱导的BV2细胞炎症损伤的病生理过程,且二者的表达存在明显正相关。Objective To explore the expressions and correlation of thrombopoietin(TPO) and interleukin-6(IL-6) in microglial cells with the intervention of lipopolysaccharide(LPS).Methods A certain concentration and correlation of LPS was given to BV2 cells,interfering for 12-24 h.There were 6 groups in the experiment.No additions for 12 h group:BV2 cells were cultured for 12 h without adding any intervention factors;LPS 0.5 mg·L^-1 for 12 h group:pre-incubated LPS solution was added to culture BV2 cells with 12 hours′ co-cultivation,resulting in a final concentration of 0.5 mg·L^-1.LPS 1.0 mg·L^-1 for 12 h group:pre-incubated LPS was added with solution to culture BV2 cells with 12 hours′ co-cultivation,resulting in a final concentration of 1.0 mg·L^-1.No additions for 24 h group:BV2 cells were cultured for 24 h,without adding any intervention factors;LPS 0.5 mg·L^-1 for 24 h group:pre-incubated LPS solution was added to culture BV2 cells with 24 hours′ co-cultivation,resulting in a final concentration of 0.5 mg·L^-1;LPS 1.0 mg·L^-1 for 24 h group:pre-incubated LPS solution was added to culture BV2 cells with 24 hours′ co-cultivation,resulting in a final concentration of 1.0 mg·L^-1.The expressions of intracellular TPO and IL-6 were detected by enzyme linked immunosorbent assay,Western blot was assayed to evaluate the expressions5 of TPO and IL-6.Results After the treatment of LPS,the expressions of protein levels of TPO and IL-6 in BV2 cells significantly increased.When 0.5 mg·L^-1 LPS had intervened the cells lasted for 12 h,TPO and IL-6 concentrations were at the peak;when the intervention of 1.0 mg·L^-1 LPS in cells lasted for 12 h,the protein levels of TPO and IL-6 showed the most significant increase,and the expressions of TPO and IL-6 showed a significant positive correlation(r=0.876,P0.01).Conclusions TPO and IL-6 are involved in the pathophysiologic process of inflammatory injury in the LPS-induced BV2 cells,and the expressions of TPO and IL-6 show obviously positive c
