摘  要:目的研究免疫相关基因HLA-G多态性与新疆哈萨克族食管癌易感性的关系。方法采用病例-对照研究方法,调查新疆哈萨克族食管癌患者132例和非食管癌患者254例,应用聚合酶链反应(PCR)技术和聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术分别对HLA-G基因14bp缺失(rs16375)和HLA-G*0105N(rs41557518)多态性进行检测,并比较2个位点不同基因型与食管癌发病风险的关系。结果 rs16375位点基因型为+14 bp/+14 bp、-14 bp/+14 bp、-14 bp/-14 bp;病例组与对照组的基因型及等位基因频率分布差异有统计学意义(χ^2=7.54,P=0.02;χ^2=6.80,P=0.01);携带-14bp/-14bp基因型的个体发生食管癌的危险性是+14 bp/+14 bp基因型的2.69倍(OR=2.69,95%CI=1.30-5.55);rs41557518位点基因型为C/-、C/C、-/-;病例组与对照组的基因型及等位基因频率分布差异无统计学意义(χ^2=1.19,P=0.28;χ^2=1.13,P=0.29);同时携带基因型-14bp/-14bp和C/C的个体发生食管癌的危险性是同时携带基因型+14bp/+14bp和C/C个体的2.82倍(OR=2.82,95%CI=1.32-6.04)。结论 HLA-G基因14bp缺失(rs16375)多态性与哈萨克族食管癌发生有关。Objective To determine the association between human leukocyte antigen G(HLA-G)polymorphisms and the susceptibility of esophageal carcinoma in Kazakh nationality in Xinjiang.Methods A case-control study was conducted.The subjects included 132 Kazakh patients with esophageal carcinoma(EC) and 254 cancer-free controls.The polymorphisms of HLA-G gene 14bp deletion at rs16375 and 0105N at rs4155751 were genotyped by polymerase chain reaction(PCR) and polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),respectively.Results There were significant differences in the frequencies of rs16375 genotype and allele between cases and controls(χ2=7.54,P=0.02;χ2=6.80,P=0.01).Compared with those who had +14bp/+14bp genotype,the individuals with-14bp/-14bp genotype had a 2.69 fold(95%confidence interval:1.30-5.55) increased risk of developing EC.There was no statistical difference in the frequencies of rs41557518 genotype and allele between cases and controls(χ2=1.19,P=0.28;χ2=1.13,P=0.29).Compared with those who had +14bp/+14bp and C/C genotypes,the individuals with-14 bp/-14 bp and C/C genotypes had a 2.82 fold(95%CI:1.32-6.04) increased risk of developing EC.Conclusion HLA-G 14bp(rs16375) polymorphism is associated with the susceptibility of esophageal cancer.HLA-G*0105N(rs41557518)polymorphism is not associated with the susceptibility of esophageal cancer in Kazakh nationality in Xinjiang.
