摘  要:目的:改进大鼠胃溃疡模型制备方法,探讨胃毒热证动物模型的制备及评价方法,为毒热证研究及清热解毒药物药理作用研究提供动物模型制备方法和检测指标选择参考。方法:将大鼠随机分为正常对照组、假手术对照组、胃溃疡模型组、清热解毒药物治疗组(溃得康组)。采取胃黏膜局部投与乙酸方法制备胃溃疡模型,治疗组术后第2d开始给药,对照组给纯水,分别于术后第1、4、8、12、18d后,取材观察胃溃疡局部改变,取血清,酶联免疫法检测血清炎症因子的含量,取组织进行病理切片,HE染色,观察黏膜炎症情况。结果:模型组大鼠胃黏膜局部溃疡,溃疡底部有附苔,溃疡周边红肿;溃疡部位组织切片病理染色可见黏膜上皮脱落,黏膜下层大量的炎症细胞浸润;血清白细胞介素-6(IL-6)和血清白细胞介素-8(IL-8)含量术后第1d开始升高,持续至12d,明显高于假手术组(P<0.01);溃得康组与模型组相比溃疡面积减小,炎症细胞浸润减少,血清IL-6、IL-8水平降低。假手术组与正常组相比,各项指标均无显著差异。结论:使用胃黏膜局部投与乙酸方法制备的胃溃疡模型急性期为毒热证胃溃疡模型,可用胃黏膜局部的溃疡状态和炎症浸润情况及血清中IL-6、IL-8含量等微观指标进行评价。Objective: In order to improve the establishment of rat's ulcer model and discuss the establishment and evaluation of animal model with stomach toxin-heat syndrome.To collect useful information for the research of heat-toxin syndrome along with heat-cleaning and toxin-resolving drugs.Methods: Rats were divided into 4 groups randomly,normal group,sham group,ulcer model group,treatment group.Applying acetic acid to certain part of the stomach mucous membrane to establish the ulcer model.Treatment group had been administrated with drugs since the second day after operation.Control group was treated with pure water.On the 1st,4th,8th,12th,18th day after operation,the healing process of ulcer was observed,the level of serum inflammatory cytokines was estimated by ELISA method.The inflammation of mucous membrane was studied through HE staining.Results: In ulcer model group,ulcer was found in the rats' stomach mucous membrane and attached musci in the bottom of the ulcer and flare around the ulcer.The sections of ulcer showed dropping mucous membrane epithlium and the severe inflammatory cell infiltration in the bottom of mucous membrane.In normal group,IL-6 and IL-8 kept rising till the 12th day after operation and there was significance between model group and normal group(P0.05).The area of ulcer in treatment group shrinked and inflammatory cell infiltration decreased,the level of IL-6 and IL-8 in serum decreased.There is no difference between normal group and sham group.Conculsion: The above-mentioned ulcer model in its acute stage can be considered as stomach heat-toxin syndrome model.Condition of the ulcer,magnitude of inflammatory cell infiltration and level of IL-6 and IL-8 in serum may be used to evaluate the stomach heat-toxin syndrome.
