摘 要:目的筛选影响鼻咽癌侵袭和转移能力的相关基因。方法利用cDNA基因芯片,比较高转移鼻咽癌细胞株5-8F和非转移鼻咽癌细胞株6-10B之间的差异表达基因,并利用在线Milano程序筛选鼻咽癌侵袭和转移相关基因,GOstat分析这些基因功能,最后荧光定量PCR鉴定差异表达基因。结果芯片分析结果显示,鼻咽癌细胞株5-8F与6-10B之间共有200多个差异表达基因。基于Milano程序分析后,发现与侵袭和转移能力相关的基因有33个,其中27个上调,6个下调。GOstat分析显示,这些基因参与了细胞移动、调节凋亡、细胞黏附等。结论经在线Milano程序分析鉴定的鼻咽癌5-8F和6-10B细胞株之间的这33个差异表达基因可能参与了鼻咽癌的侵袭和转移。Objective To screen potential genes associated with invasion and metastasis in nasopharyngeal carcinoma. Methods cDNA microarray was used to detect the differential expression in 5-8F cells with the ability of high metastasis and 6-10B cells lacking the metastatic ability. Subsequently,genes of invasive and metastasis were screened by Milano software tool. The function of these genes was analyzed by GOstat program. And real-time PCR was utilized to confirm the reliability of differentially expressed genes. Results More than 200 genes were identified to have the differential expression. After the analysis with Milano software tool ,33 genes were found to be likely associated with cell invasion and metastasis. Among these genes,27 genes were showed to be upregulated, and 6 genes were indicated to be downregulated. Finally,the reliability of differentially expressed genes was confirmed by real-time PCR. Conclusion Differentially expressed genes shown in NPC 5-8F cells eompared to 6-10B cells with the identifieation of online Milano program analysis were likely associated with the invasion and metastasis of NPC cells.
