基质金属蛋白酶8在食管鳞癌中的表达及其临床意义

摘　　要：目的探讨食管鳞状细胞癌中基质金属蛋白酶-8(matrix metalloproteinase 8,MMP-8)的表达及其与肿瘤浸润、转移的关系。方法实时荧光定量PCR方法检测67例食管鳞癌组织和27例远癌组织中MMP-8 mRNA的表达;Western blot法检测其中15例食管鳞癌组织及其配对的15例远癌组织中MMP-8蛋白的表达,统计分析MMP-8表达水平与食管鳞癌的临床分期以及浸润转移的关系。结果①MMP-8在食管鳞癌组织和远癌组织中均有不同程度的表达,在癌组织的表达水平显著低于远癌组织(mRNA表达水平P=0.003,蛋白表达水平P=0.031)。②T1、T2、T3、T4期食管鳞癌组织中MMP-8 mRNA表达存在显著差异(P=0.002),MMP-8表达随着肿瘤浸润程度的加深而升高;MMP-8蛋白表达量在T1～T4期中也呈增加的趋势。③MMP-8 mRNA和蛋白表达水平在食管鳞癌的不同淋巴结转移组之间没有显著差异。结论 MMP-8在食管鳞癌组织中低表达,但又随肿瘤浸润加深而表达升高,可能反映了MMP-8在食管鳞癌发生和浸润中的不同作用。Objective To study the expression of matrix metalloproteinase 8（MMP-8） in esophageal squamous cell carcinoma（ESCC） and its relationship with tumor invasion and metastasis.Methods MMP-8 mRNA expressions in 67 ESCC tissue specimens and in 27 normal tissue specimens were detected by real-time PCR.MMP-8 protein expression in 15 paired specimens of ESCC and in normal tissues were detected by Western blotting.The relationship between MMP-8 expression level and the clinical stage,invasion and metastasis of ESCC was statistically analyzed.Results MMP-8 expression was detected in both ESCC tissues and normal tissues,and the expression level in ESCC tissues was significantly lower than that in normal tissues（P=0.003 for mRNA expression and P=0.031 for protein expression）.MMP-8 expression in ESCC tissues had significant differences among T1,T2,T3 and T4 stages（P=0.002）,increasing from T1 to T4 stage,and rose when tumor invasion degree heightened.No obvious correlation was found between the MMP-8 mRNA and protein expression levels and lymph node metastasis of ESCC.Conclusions Expression of MMP-8 is low in ESCC tissues and increases with elevated invasion degree,which may reflect different mechanisms of MMP-8 in ESCC carcinogenesis and invasion.
