摘 要:目的观察Intermedin1-53(IMD1-53)对两肾一夹(2-kid-ney1-clip,2K1C)高血压大鼠的影响作用及其机制。方法采用2K1C复制肾性高血压大鼠模型;检测心功能,免疫组化法检测心脏IMD的水平;放射免疫学方法检测血浆中IMD和血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)的含量;采用硝酸还原酶法测定血清中一氧化氮(nitric oxide,NO)含量,Griess化学比色法测定血清中一氧化氮合酶(nitric oxidesythase,NOS)活性。结果 2K1C高血压大鼠心功能抑制,心脏IMD的水平及血浆中IMD和AngⅡ的含量升高,血清中NO、NOS的含量降低;与2K1C组比IMD1-53组大鼠心功能明显改善,血浆中AngⅡ的含量降低,血清中NO和NOS的含量升高,差异均有显著性。结论 IMD在2K1C时表达升高,外源性IMD具有明显降低肾性高血压的作用,其降压作用机制可能与增加NO/NOS有关。Aim To observe the effect of Intermedin1-53(IMD1-53)on two-kidney-one-clip(2K1C) hypertensive rats and its mechanism.Methods The renal hypertensive rats of 2K1C was adopted to detect their heart function.The IMD level of heart was determined by immunohistochemisty;the content of IMD and angiotensin Ⅱ in plasma was detected by radioimmunosaasy methods;the content of nitric oxide(NO) in serum was measured by nitric acid reductase method,and Griess chemical colorimetry technique was used to test the activity of nitric oxide synthase(NOS) in serum.Results The heart function of 2K1C hypertensive rats was inhibited.The IMD level heart and the content of IMD and Ang II in plasma increased,while the content of NO and NOS in serum decreased.Compared with 2K1C group,the rat heart function of IMD1-53 group was significantly improved,with decreased content of AngⅡin plasma and increased content of NO and NOS in serum.The differences were obvious.Conclusions IMD expression increased in 2K1C;exogenous IMD has the significant function of decreasing renal hypertension;its mechanism has something to do with the increasing of NO/NOS.
