摘 要:目的:考察蒿甲醚与其抗肿瘤增效剂—亚铁盐的配伍稳定性,为临床合理用药提供依据。方法:采用UPLC法检测蒿甲醚分别与游离亚铁离子(硫酸亚铁)及络合亚铁离子(EDTA亚铁)在高温(60℃)下共存含量随时间的变化。结果:在弱酸至碱性环境下,pH值对蒿甲醚的稳定性几无影响。当蒿甲醚与游离亚铁离子共存时,其降解速率随亚铁离子浓度增大而增加,直至硫酸亚铁与蒿甲醚的质量比为1∶1时,降解速率增加约1倍,达最大值,此后不再随亚铁离子浓度增大而增加。络合亚铁离子对蒿甲醚降解速率的影响与游离亚铁离子相似。结论:两种形式的亚铁离子对蒿甲醚的稳定性均有一定影响,合用时应尽量避免二者长时间共存。OBJECTIVE To investigate the compatibility of artemether and its anti-tumor synergist-ferrous salt for rational drug use. METHODS UPLC method was used to determine the variations in concentration of artemether coexisted with free ferrous ion (ferrous sulfate) and chelating ferrous ion (ferrous EDTA), respectively under high temperature (60℃). RESULTS The effect of pH on the stability of artemether was not significant, while free ferrous ion could decrease the stability. Degra dation rate of artemether increased with ferrous ion concentration and reaehed the maximum (in ahout double time) until the mass ratio of ferrous sulfate to artemether was 1 : 1. The effect of chelating ferrous ion on degradation rate of artemether was similar to that of free ferrous ion. CONCLUSION Both free ferrous ion and chelating ferrous ion have an effect on the stability of artemether. Artemether and ferrous salt should avoid coexistence for a long time.
