摘  要:利用番茄内质网小分子热激蛋白(ERsHSP)特异性抗体,对番茄果实蛋白进行Western分析,以测定低温冷藏下番茄果实中ERsHSP的表达量,并测定果实硬度、腐烂度和失重率等指标,以比较中蔬4号转ERsHSP基因番茄和未转基因番茄果实在4℃低温下的耐冷藏性.结果表明:在4℃冷藏30 d期间,转基因番茄果实具有较高的ERsHSP表达水平,而未转基因番茄果实中没有ERsHSP的表达;相对于未转基因番茄果实,转基因番茄果实冷害症状轻,并具有较高的果实硬度(平均值为2.84 kg/cm2)、较低的果实腐烂度(平均值为21.03%)和失重率(平均值为6.33%).In order to detecting the expression level of ERsHSP in tomato fruits under low temperature(4℃),total proteins of fruits were subjected to Western analysis with special polyclonal antibody against ERsHSP.To analysis the chilling tolerance of ERsHSP transgenic tomato fruits and non-transgenic tomato fruits(Lycopersicon esculentum var.Zhongshu 4) under low temperature,the fruit hardness,decay percentage and weight loss was measured.The results indicated that during the whole cold storage,there were high levels of ER-sHSP expression in transgenic tomato fruits,but no ER-sHSP protein was detected in non-transgenic tomato fruits.As a result,the chilling tolerance of transgenic tomato fruits with ER-sHSP over-expression was significantly improved.The statistical data indicated that the transgenic tomato fruits,in comparison,have higher hardness(with an average value 2.84 kg/cm2),lower percentage of decay(with an average value 21.03%) and less loss of weight(with an average value 6.33%).
