小麦蛋白膜的体外生物相容性研究

摘  要:目的研究小麦蛋白的体外生物相容性,探索其用作生物医用材料的潜在可能。方法以聚乳酸膜为对照,进行了细胞相容性试验(包括浸提液细胞毒性试验和细胞直接接触试验)和血液相容性试验(包括溶血试验、血小板粘附试验和动态凝血时间试验)。结果小麦蛋白膜浸提液细胞毒性评价合格;大鼠成骨样细胞Ros17/2.8在小麦蛋白膜上的粘附和铺展程度比在聚乳酸膜上更充分;小麦蛋白膜溶血率为0.5%,符合医用材料对溶血控制的要求,血小板粘附于小麦蛋白膜后的形态指数为1.56,低于聚乳酸组的2.54,即激活程度更低,血液在小麦蛋白膜表面的初凝时间为14 min,较聚乳酸组延迟了9min。结论小麦蛋白具有良好的生物相容性,可望用作生物医用材料。Objective To explore the potential of wheat gluten being used as biomedical material by studying its biocompatibility in vitro. Methods Cytocompatibility including cytotoxicity of gluten film extracts and cell co-culture experiments,as well as haemocompatibility including haemolytic test,platelet adhesion test and dynamic clotting time test were carried out in vitro with L-polylactide(PLA) as control.Results The cytotoxicity evaluation was acceptable.Osteo-like cells Ros17/2.8 attached and spread much better on gluten than on PLA.The hemolytic ratio of gluten was 0.5% indicating no obvious hemolytic influence.The deformation index of platelets adhered on gluten was 1.56,less than that on PLA(2.54) indicating lower degree of platelet activation.The initial clotting time of gluten was 14 min,9 min longer than that of PLA group indicating better antithrombotic ability.Conclusion Wheat gluten might have great potential as biomedical materials.
