摘  要:本研究探讨多发性骨髓瘤(MM)患者骨髓间充质干细胞(MSC)趋化相关因子HGF、SDF-1、MCP-1和IL-8的基因表达水平及临床意义。应用实时定量RT-PCR方法检测20例初发多发性骨髓瘤患者骨髓间充质干细胞HGF、SDF-1、MCP-1和IL-8的表达水平。结果显示,HGF mRNA在MMMSC的表达明显高于对照组,具有统计学意义(p<0.01),SDF-1 mRNA的表达较对照组低(p<0.05),MCP-1和IL-8 mRNA的表达在两组间无统计学差异(p>0.05)。结论:MM患者骨髓MSC均表达HGF、SDF-1、MCP-1和IL-8,MM患者骨髓微环境中MSC趋化相关因子表达存在异常。This study was aimed to investigate the mRNA expression levels of hepatocyte growth factor(HGF),stromal cell-derived factor-1(SDF-1),monocyte chemotactic protein-1(MCP-1) and interleukin-8(IL-8)in bone marrow mesenchymal stem cells(MSC) from multiple myeloma(MM) patients.The mRNA expression levels of HGF,SDF-1,MCP-1 and IL-8 in bone marrow MSC from 20 newly diagnosed MM patients were detected by real time quantitative RT-PCR and were compared with that in 9 controls.The results indicated that the mean mRNA expression level of HGF was up-regulated in MM patients,as compared with controls(p0.01).However,the mean mRNA expression level of SDF-1 mRNA was down-regulated in MM patients,as compared with controls(p0.05).There was no significant difference in the mRNA expression levels of MCP-1 and IL-8 between MM and control cohorts(p0.05).It is concluded that BM-MSC from MM patients express HGF,SDF-1,MCP-1,IL-8,but these chemotaxis-related factors expression of bone marrow microenvironment cellular component are dysregulated in MM patients,which may result from the interplay between MM cells and MSC.
