摘  要:目的研究20 nm银纳米颗粒(AgNPs)对脑胶质瘤C6细胞磁流体热疗的作用。方法脑胶质瘤C6细胞分为对照组、AgNPs组、磁流体热疗组和AgNPs结合磁流体热疗组。通过集落形成实验检测20 nm AgNPs对脑胶质瘤C6细胞热疗的效果;流式细胞术检测AgNPs对细胞周期和凋亡率的影响;HE染色检测AgNPs对荷瘤鼠磁流体热疗凋亡的影响。结果与对照组比较,20 nmAgNPs能够引起脑胶质瘤C6细胞G2/M期阻滞,并使细胞在接受磁流体热疗后凋亡率显著提高(P<0.05)。结论银纳米颗粒能提高脑胶质瘤C6细胞的热敏感性。Objective To investigate the effect of 20 nm silver nanoparticles(AgNPs) on brain glioma C6 cells after magnetic fluid hyperthermia.Methods Brain glioma C6 cells were divided into four groups of control,AgNPs,magnetic fluid hyperthermia and AgNPs combined with magnetic fluid hyperthermia.The effect of 20 nm AgNPs on thermotherapy of glioma C6 cells was detected by colony-forming assay.The cell cycle and apoptosis after irritation were tested by flow cytometry.HE staining was used to detect the effect of AgNPs on apoptosis in tumor-bearing mice after magnetic fluid hyperthermia.Results Compared with control group,20 nm AgNPs could induce the arrest of cell cycle in G2/M phase and promote cell apoptosis after magnetic fluid hyperthermia (P0.05).Conclusion The nanoparticles with core-shell structures of silver and iron can significantly enhance thermosensitivity of brain glioma C6 cells.
