摘 要:目的探讨川芎嗪(TMP)对阿尔茨海默病(AD)模型小鼠脑胆碱乙酰基转移酶(ChAT)和N-甲基-D-天冬氨酸(NMDA)受体的影响。方法 56只小鼠随机分为五组:正常对照组(A组,10只)、D-半乳糖(D-gal)模型组(B组,10只)、TMP低、高剂量组(C、D组,各12只)和哈伯因阳性对照组(E组,12只)。Morris水迷宫测试学习记忆功能,同时进行大脑皮层、海马ChAT活性测定和NMDA受体结合试验。结果与A组相比,B组学习记忆能力和海马NMDA受体含量均下降(P<0.01);TMP可明显改善模型小鼠的学习记忆能力,增强皮层和海马ChAT活性,上调海马NMDA受体数目(P<0.05)。结论 TMP可明显提高模型小鼠的学习记忆功能,这可能与增强ChAT活性及保护NMDA受体有关。Objective To explore the effect of tetramethylpyrazine(TMP) on the choline acetyltransferase(ChAT) activity and N-methyl-D-aspartate(NMDA) receptor in Alzheimer′s disease(AD) model mice.Methods Fifty-six mice were divided into 5 groups of A(normal control group,10 mice),B(D-galactose model group,10 mice),C(low-dose TMP group,12 mice),D(high-dose TMP group,12 mice) and E(huperzine positive control group,12 mice).Learning and memory ability was tested by Morris water maze.ChAT activity,the binding sites(Bmax) and the affinity(KD) of NMDA receptor in the cerebral cortex and hippocampus were determined.Results Compared with group A,the learning and memory dysfunction and NMDA receptor binding sites in the hippocampus were all decreased in group B(P0.05).TMP markedly improved cognitive dysfunction,enhanced ChAT activity in the cerebral cortex and hippocampus and NMDA receptor binding sites in the hippocampus of mice treated with D-galactose(P0.05).Conclusion TMP can significantly enhance the learning and memory ability in AD model mice,probably through promoting ChAT activity and protecting NMDA receptor.
