摘  要:以永生化山羊子宫内膜上皮细胞作为体外模型,采用革兰氏染色法观察乳酸菌和致病菌对子宫内膜上皮细胞的黏附特性;通过平板计数法观察乳酸菌对致病菌的黏附拮抗作用。结果显示,嗜酸乳杆菌、粪肠球菌、盲肠肠球菌对子宫内膜上皮细胞均表现出很强的黏附特性,黏附细菌数分别为每个细胞87、45、34个。黏附拮抗试验显示,3株乳酸菌均使致病菌的黏附率显著降低。其中嗜酸乳杆菌对大肠杆菌的黏附拮抗作用最强,使C83903的黏附率由38.33%下降至9.95%,ATCC25922的黏附率由65.42%下降至8.27%。粪肠球菌和盲肠肠球菌对金黄色葡萄球菌的黏附率由86.18%分别下降至8.71%、8.63%。证实永生化山羊子宫内膜上皮细胞作为细菌黏附性能研究的体外模型是有效可行的。Using immortalized endometrial epithelial cells(EEC) as an in vitro model,the adherence properties of lactic acid bacteria(LAB) and pathogenic bacteria to EEC were observed by Gram's staining,while their antagonistic activity against pathogenic bacteria was observed by plate counting.All LAB strains showed adhesion to EEC,being 87/cell for Lactobacillus acidophilus,45/cell for Enterococcus faecalis,and 34/cell for Enterococcus cecorum.The relationship between adhesion and acid-forming of LAB was observed,and acid environment increased the adhesion ability of bacteria.In co-culture assays,a significant decrease in the number of adhering pathogenic bacteria was observed.L.acidophilus showed antagonistic activity against Escherichia coli,the adherence rate of C83903 decreased from 38.33% to 9.95% and ATCC25922 from 65.42% to 8.27%.The adherence rate of Staphylococcus aureus decreased from 86.18% to 8.63% and from 86.18% to 8.71% when it was co-cultured with E.cecorum and E.faecalis,respectively.In conclusion,these findings suggest that the potential usefulness of immortalized hircine endometrial epithelial cells as a new in vitro model for studying the adherence properties of bacteria.
