摘 要:目的:探讨以O/W型微乳液为溶剂提取当归的可行性和适宜性。方法:以O/W型微乳为溶剂提取当归,采用不同溶剂(微乳、不同浓度乙醇、水)进行不同提取方法(加热、微波、超声波)的提取实验,用HPLC测定当归微乳提取液中阿魏酸的含量,比较微乳对当归中阿魏酸的提取效率及其影响因素。采用薄层色谱法比较微乳与其他溶剂提取当归中脂溶性和水溶性成分的异同。结果:用微乳加热提取当归,可提高阿魏酸的提取率,提取效果与50%乙醇相当;微乳的配方组成和提取方法对当归中阿魏酸的提取具有明显影响;同时薄层色谱结果表明,微乳提取液不仅可以增加水溶性酚酸类成分的溶出,还可以增加挥发油等脂溶性成分的溶出。结论:利用微乳的特性,可同时提取中药中不同溶解性质的成分,符合中医临床多组分用药的特点。可为微乳用于中药提取的可行性和适宜性提供实验依据。Objective:To investigate the feasibility of extraction of the compounds with moderate polarity from Angelica sinensis with O/W microemulsion.Methods:O/W microemulsion,water and ethanol were used as the solvents to extract Angelica sinensis.The content of ferulic acid in extracts of Angelica sinensis was determined by HPLC.We compared the efficiency of extracting Angelica sinensis by using the different media,different formulations of O/W microemulsion.We used TLC to compare the differences of Angelica sinensis' extracts by different extraction methods.Results:The extracting yield of ferulic acid was improved by using microemulsion,which was similar to 50% ethanol.The formulations of microemulsion and extracting methods remarkably affected the extraction of ferulic acid from Angelica sinensis.TLC results indicated that microemulsion extracts not only increased the solubility of ferulic acid,but also improved the solubility of fat-soluble compounds.Conclusion:O/W microemulsion is feasible to extract Angelica sinensis as a solvent,and meets the demand of clinical application for TCM.It not only has the advantage of extracting the polarity compound,but also avoids the usage of organic solvents,saves energy and reduces the links of manufacture.These are beneficial to the sustained development and low-carbon economy of Chinese Materia Medica.
