摘 要:目的检测胃癌组织和癌旁组织中肿瘤干细胞标志物CD133的表达,探讨其与临床病理参数的相关性和在肿瘤诊断中的意义。方法收集36例胃癌患者手术切除的癌组织和对应的癌旁组织,用RT-PCR检测CD133的mRNA表达,用免疫组织化学技术检测CD133蛋白质的表达,分析CD133 mRNA在胃癌组织中的表达与临床病理参数的相关性。结果 CD133 mRNA表达阳性率为胃癌组织66.7%(24/36),癌旁组织为16.7%(6/36),差异有统计学意义(P<0.01);胃癌组织中CD133表达阳性率与肿瘤分化状态相关,低、未分化组高于中、高分化组(P<0.01),但与年龄、性别、肿瘤大小、浸润深度、TNM分期和有无淋巴结转移无关。结论肿瘤干细胞标志物CD133的表达与胃癌的发生及分化状态相关,可作为胃癌诊断的一个新的分子标志物。Objective To detect the expression of cancer stem cell marker CD133 in paired tumor and non-cancerous tissues of gastric cancer(GC) patients and explore correlation of CD133 expression with the clinicopathological parameters of GC and the significance for diagnosis of GC.Methods The surgically resected paired cancerous and matching adjacent non-cancerous tissue of from 36 GC patients were collected.The expression of CD133mRNA was examined by reverse transcription-PCR(RT-PCR).Immunohistochemistry(IHC) was employed to detect CD133 protein level was detected by immunohistochemistry.Correlation between CD133 expression and clinicopathological parameters was analyzed.Results The positive rate of CD133mRNA expression in GC tissues was 66.7%,which was significantly higher than that in the matching,adjacent non-cancerous tissues(16.7%,P0.01).CD133 expression correlated with the differentiation stage of tumor.The positive rates of CD133 in poorly differentiated and undifferentiated GC tissues were significantly higher than those in the moderately and well-differentiated GC tissues(P0.05),but no association was found between CD133 expression and other clinicopathological parameters,including age,gender,tumor size,TNM stage,depth of invasion,and lymph node metastasis.Conclusions The tumorigenesis of GC may correlate with cancer stem cell.The expression of CD133,a marker of cancer stem cell,associated with the stage of differentiation of GC,so it may serve as a novel diagnostic marker for human GC.
