摘  要:目的:探讨HPV感染及k-ras基因突变与直肠腺癌的关系。方法:应用PCR/SSCP银染法检测75例直肠腺癌患者癌及相应正常大肠黏膜组织中HPV DNA和k-ars基因第12、13密码子。结果:直肠腺癌组织中HPVDNA阳性检出率为73.3%(55/75),而正常黏膜未检测到HPV DNA。HPV感染与患者年龄、Duke’s分期及淋巴结转移有关,而与患者的族别、性别、肿瘤直径、大体类型、组织学类型及浸润深度无关。75例直肠腺癌组织中检测到k-ras基因突变34例(45.3%),均与患者年龄、肿瘤的大体类型、组织学类型、侵袭深度、淋巴结转移及Duke’s分期无关,且与HPV感染亦无关联(χ2=0.240,P=0.624,rP=0.056)。结论:HPV感染可能是导致直肠腺癌恶性表型的重要因素;而k-ras基因突变与HPV感染无关。Aim:To explore the relationship between HPV infection,k-ras gene mutation and rectal adenocarcinoma.Methods:The HPV DNA, the code 12,13 of k-ras in 75 cases of rectal adenocarcinoma tissue and the corresponding normal tissue were detected respectively by PCR and PCR-SSCP.Results:55 out of 75(73.3%) cases showed positive HPV expression while normal tissue showed negative expression. HPV infection was related to the age of patients,Duke’s stage and lymph node metastasis,but not to the ethnic of patients,gender, tumor size,gross type,histological type or the depth of invasion.34(45.3%) cases showed k-ras gene mutation in all 75 cases,which had no relation to the indexes mentioned above,and the k-ras gene mutation had no assoication with the HPV infection(χ2=0.240,P=0.624,rP=0.056).Conclusion:HPV infection may play an important role in the development of premalignant lesions leading to the neoplastic phenotype,and k-ras gene mutational may be a common and early event in colon carcinogenesis.
