淮南市不同生境中主要尘螨变应原的基因检测

摘  要:目的检测3种不同生境中尘螨4种变应原。方法于2009年6—8月在淮南地区仓储、居室及公共环境采集样本各40份,提取样本总RNA,RT-PCR合成cDNA,应用PCR特异性扩增Der f 1、Der f 2、Der p 1、Der p 2,并对不同生境中变应原检测结果进行统计学分析。结果从尘螨基因组中特异扩增出大小约646、4559、23、408 bp的基因片段。仓储环境和居室环境中尘螨变应原的检测阳性率均高于公共环境(P<0.05),仓储环境中粉尘螨变应原阳性率较高(Der f 1为65.0%,Der f 2为62.5%),居室环境中屋尘螨变应原阳性率较高(Der p 1为80.0%,Der p 2为62.5%)。结论不同生境中不同尘螨变应原具有较大差异,可能与温湿度差异及尘螨的食性、习性有关。对尘螨基因组进行RT-PCR及PCR检测是简便有效的尘螨变应原检测方法。Objective To detect gene encoding house dust mite allergens of 1,2 group from three different habitats by RT-PCR and PCR.Methods One hundred and twenty dust samples were collected from storage,dwelling and public circumstance in Huainan city from June to August in 2009.The total RNA was extracted firstly,and RT-PCR was used to acquire cDNA subsequently.Then PCR was performed to specifically amplify Der f1,Der f2,Der p1,Der p2,and results were analyzed by statistical treatment.Results Four gene sequences(646 bp,455 bp,923 bp,408 bp) were specifically amplified from house dust mite genome.The positive ratios of house dust mite allergens in storage habit and dwelling habit were significantly higher than those in public circumstance,and the positive ratio of Der f1,Der f2 in storage habit and Der p1,Derp2 in dwelling habit were even higher respectively.Conclusion Various house dust mite allergens from three habitats are significantly different,and the important factors may be the variation of temperature and humidity,habit and food habit of house dust mites.Furthermore,RT-PCR and by PCR for the detection of house dust mite genome should be the convenient and effective methods.
