摘  要:目的:观察Delta阿片受体激动剂DADLE([D-Ala2,D-Leu5]enkephalin)对大鼠原代皮层神经元转录的影响。方法:体外培养皮层神经元,免疫荧光法鉴定其纯度,以不同浓度的DADLE与神经元共培养不同的时间,应用[3H]标记脲嘧啶掺入法测定神经元的转录水平,MTT法检测细胞活力。结果:DADLE在不同浓度均能抑制神经元的转录,但是其抑制率与浓度不呈线性关系,当浓度增加到1nmol以上时其抑制率绝对值稳定在40%~60%左右。DADLE的转录抑制率与作用时间也不呈线性关系,当时间超过24h后,其抑制率绝对值维持在40%~60%。将DADLE洗脱72h后其抑制效果完全消失。MTT检测显示各个浓度的DADLE对神经元的活力影响无显著差异。结论:DADLE能可逆、无损伤抑制神经元的转录。Objective To explore the effect of delta opioid agonist [D-Ala2,D-Leu5] enkephalin (DADLE) on the primary cortical neuronal transcription in rats. Methods Primary rat cortical neurons were cultured in vitro and the purity of the cells were determined by immunofluorescence assay. DADLE of different concentrations was co-cultured with the cells for different time. The transcript levels of the neurons were detected by [3H]-uracil incorporation and the neuronal viability was measured using MTT assay. Results DADLE of different concentrations inhibited neuronal transcription but the inhibition rate was not linearly related with the concentration and the absolute value of the rate maitained at 40% ~ 60% as the concentration increased to 1nM or above. Also, the inhibition rate was not linearly related with time and the absolute value of the rate maitained at 40% ~ 60% after 24 hours of action. The inhibition effect vanished 72 hours after elution of DADLE. The effect on the neuronal viability did not differ significantly among DADLE at different concentrations. Conclusions DADLE reversibly and harmlessly inhibits neuronal transcription.
