摘  要:合成了5′-硝基水杨基荧光酮(5′-NSF),并制备了与钼(Ⅵ)所形成的结合比为[n5′-NSF∶n钼(Ⅵ)]1比1的配合物作为新型的光度探针。应用紫外-可见分光光度法研究了5′-NSF-钼(Ⅵ)配合物与蛋白质的相互作用。结果表明:在pH3.6盐酸-乙酸钠缓冲溶液中,5′-NSF-钼(Ⅵ)配合物的吸收峰位于532 nm波长处,且其吸光度因加入适量的OP而得到增加。但因与蛋白质之间的相互作用,引起吸收强度降低。且其吸光度的降低值ΔA与蛋白质浓度,以BSA和HSA为例,分别在1.0~35.0,1.0~40.0 mg.L-1之间呈线性关系;表观摩尔吸光率依次为1.02×106,1.03×106L.mol-1.cm-1。该方法用于尿液、奶粉和人血清中蛋白质的测定,回收率在92.8%~96.0%之间,相对标准偏差(n=5)小于2.0%。5′-Nitro-salicyfluorone(5′-NSF) was synthesized,and its coordination complex with Mo(Ⅵ) with binding ratio [n5′-NSF∶nMo(Ⅵ)] of 1 to 1 was prepared which was used as a new photometric probe.Mechanism of the interaction of this complex with protein was studied by UV-VIS spectrophotometry.It was found that the absorption maximum of coordination complex of 5′-NSF-Mo(Ⅵ) lies at the wavelength of 532 nm in an acetate buffer medium of pH 3.6,and that the absorption intensity was enhanced by the presence of appropriate amount of OP.However,due to the interaction of this coordination complex with protein,the absorption intensity was deceased,and the magnitude of decrease in absorbance(ΔA) was found to keep linear relationship with mass concentration of protein in the ranges of 1.0 to 35.0 and 1.0 to 40.0 mg·L-1 for BSA and HSA and with values of molar absorptivity 1.02×106 and 1.03×106L·mol-1·cm-1 respectively.The proposed method was used in the determination of protein in samples of urine,milk powder and human serum,giving values of recovery in the range of 92.8%-96.0% and RSD′s(n=5) less than 2.0% respectively.
