组织芯片检测舌鳞状细胞癌组织中KLF6、p21及c-Jun的表达

摘　　要：目的探讨舌癌和正常舌组织中核转录因子KLF6、p21及c-Jun蛋白的表达、其相关性及临床意义。方法应用高通量组织芯片技术结合免疫组织化学法,检测包含人类舌鳞状细胞癌组织、舌良性肿瘤组织、因良性病变而活检的正常舌组织的组织芯片中KLF6、p21及c-Jun的表达情况,结合临床病理资料进行统计分析。结果舌癌组织的KLF6表达明显低于正常舌组织和舌良性肿瘤组织,KLF6在低分化舌癌中的阳性表达率100.00%(4/4)明显高于高分化的66.67%(8/12)和中分化的50.00%(2/4);舌癌组织的p21表达明显低于正常舌组织和舌良性肿瘤组织,p21在淋巴结未转移组和临床Ⅰ、Ⅱ期组舌癌组织的表达(9/10)明显高于淋巴结转移组和临床Ⅲ、Ⅳ期组(5/10);舌癌组织的c-Jun蛋白表达显著高于正常舌组织和舌良性肿瘤组织(P<0.05),c-Jun在淋巴结未转移组和临床Ⅰ、Ⅱ期组舌癌组织的表达(8/10)明显低于淋巴结转移组和临床Ⅲ、Ⅳ组(10/10)。KLF6与c-Jun表达存在正相关(P<0.05);c-Jun与p21的表达也存在正相关(P<0.05);11例癌组织的KLF6与p21同为阳性表达,3例癌组织二者同为阴性表达,但KLF6与p21蛋白的表达相关不显著(P>0.05)。结论 KLF6、p21和c-Jun三者与舌癌的发生发展有关,联合检测3个指标,在病理学诊断上有重要的参考价值,可辅助舌癌的临床早期诊断、鉴别诊断及预后判断。【Objective】 To detect the expression levels of KLF6,P21 and c-Jun in tongue squamous cell carcinoma and normal tongue tissue,and to assess their correlations and clinical significance.【Methods】 High flux tissue chip technology combined with immunohistochemistry were used to test the expression levels of KLF6,p21 and c-Jun in 24 cases of various tongue tissues（21 cases of tongue squamous cell carcinoma,1 case of hyperplasia,2 cases of normal tongue tissue）.【Results】 The expression of KLF6 and p21 in tongue squamous cell carcinomas was obviously lower than that in hyperplastic tissue and normal tissues.The positive expression rate of KLF6 in poorly-differentiated tongue cancers was higher than those in well-differentiated and moderately-differentiated tongue cancers.The expression level of p21 in non-metastasis group and early clinical period was obviously higher than that in metastasis group and late clinical period.The expression of c-Jun in tongue cancers was higher than that in normal tongue tissues（P〈 0.05）.And the expression of c-Jun in non-metastasis group and early clinical period was obviously lower than that in metastasis group and late clinical period.Furthermore,there were positive correlations between KLF6 and c-Jun（P〈 0.05）,and p21 and c-Jun（P 〈0.05）.However,there was no correlation between KLF6 and p21（P 〈0.05）.In 11 cancer cases,the expressions of both KLF6 and p21 were positive,but in 3 cancer cases,both were negative.【Conclusion】 Our results indicate that there are positive correlations among the development of tongue squamous cell carcinoma and KLF6,p21 and c-Jun.The combined immunohistochemical examination on KLF6,p21 and c-Jun has significant value in the pathological diagnosis,differential diagnosis and prognosis of tongue squamous cell carcinoma.
