摘  要:目的在前期实验的基础上,通过观察对比大鼠48 h快速动眼(REM)期睡眠剥夺(SD)和6 h睡眠恢复(SR)及莫达非尼干预后大鼠下丘脑orexin A神经元的表达,进一步探讨莫达非尼对睡眠的影响作用。方法成年雄性Sprague-Dawley大鼠20只,随机数字表法分为SD组10只和SR组10只,每组又分为用药组(drug group,DG)5只和非用药组(non-drug group,NDG)5只。采用小平台水环境法建立大鼠REM期SD模型。SD组剥夺睡眠48 h,SR组剥夺睡眠48 h后再恢复睡眠6 h。DG大鼠每日予莫达非尼混悬液灌胃给药,NDG同样时间予梭甲基纤维素钠(CMC)灌胃。到时间点后立即灌注处死,免疫组织化学方法观察大鼠下丘脑orexin A阳性神经元的数量。结果在SD组中,DG下丘脑orexin A阳性神经元的数量明显高于NDG(P<0.05);而在SR组中,DG与NDG之间差别不明显(P>0.05)。结论莫达非尼可能是通过活化下丘脑促觉醒肽orexin A的分泌和表达实现促觉醒作用。【Objective】 To observe the expressions of orexin A neurons in rat's hypothalamus after rapid eye movement(REM) sleep deprivation,revival and modafinil intervention and to further define modafinil's functions.【Methods】 Twenty male Sprague-Dawley rats were used to establish rat model of REM sleep deprivation by the small platform method.They were divided into sleep deprivation group(SDG,10) and revival group(SRG,10) at random.Each one contained drug group(DG,5) and non-drug group(NDG,5).The rats in SDG were deprived sleep for 48 hours and those in SRG were deprived sleep for 48 hours then recovered sleep for 6 hours.The rats in DG were perfused with modafinil suspension and those in NDG were done with sodium carboxymethylcellulose.They were put to death by perfusion at the point-in-time.Immunohistochemical method was performed to observe total number of orexin A positive neurons in rat's hypothalamus.【Results】 Within SDG,the number of orexin A positive neurons in rat's hypothalamus of DG was larger than the one of NDG(P〈 0.05);but within SRG,there was no significant difference between them(P 〉0.05).【Conclusion】 Modafinil realizes its arousal function maybe by activating the secretion and expression of orexin A in hypothalamus.
