CXCL12及其受体CXCR4在喉鳞癌中的表达及其临床意义

摘　　要：目的探讨CXCL12以及CXCR4基因在喉鳞状细胞癌中的表达及其临床意义。方法取39例喉鳞癌及28例正常喉组织,应用半定量的RT-PCR方法检测CXCL12以及CXCR4基因mRNA的表达。结果正常喉黏膜CXCL12基因mRNA表达量为(1.22±0.41),在有淋巴结转移(N1,N2),晚期分级(T3,T4)的喉癌组织中CXCL12表达量升高,其mRNA值分别为(1.63±0.47)和(1.57±0.61),与正常喉黏膜组织相比差异有统计学意义(P<0.05)。正常喉黏膜组织的CXCR4基因mRNA表达量为(0.98±1.02),CXCR4基因mR-NA在喉癌中表达与淋巴结转移、T分级及病理分化无关,与正常喉黏膜组织mRNA表达相比,其差异没有统计学意义(P>0.05)。结论与CXCR4基因比较,CXCL12基因表达与喉癌发展关系更为密切,CXCL12基因mRNA表达升高可能与喉鳞状细胞癌的淋巴结转移及演进有关。【Objectives】To detect the mRNA expression of CXCL12 and CXCR4 genes in laryngeal squamous cell carcinoma,and to explore their clinical significances.【Methods】39 Fresh laryngeal cancer samples and 28 cases of normal control laryngeal tissues were examined with semi-quantitative reverse transcription polymerase chain reaction（RT-PCR）.【Results】The CXCL12 mRNA expression of normal laryngeal tissues were 1.22±0.41.The CXCL12 mRNA expression in the laryngeal squamous carcinoma with lymph node metastasis and T3,T4 stage were 1.63±0.47 and 1.57±0.61,respectively.The mRNA expression in the laryngeal squamous carcinoma with lymph node metastasis and T3,T4 stage were higher than that of normal laryngeal tissues,and the differences were significant in statistics（P 0.05）.The CXCR4 mRNA expression of normal laryngeal tissues were 0.98±1.02.The CXCR4 mRNA expression had no relationship with lymph node metastsis,T stage and the pathology differentiation of laryngeal carcinoma.The differences of CXCR4 mRNA expression between normal laryngeal tissues and laryngeal squamous carcinoma had no significance（P 0.05）.【Conclusions】CXCL12 expression was related to a greater degree to laryngeal carcinoma development,compared with CXCR4 expression.Evaluation of CXCL12 expression is useful for determining tumor properties,including nodal metastasis and progression in patients with laryngeal squamous cell carcinoma.
