摘  要:目的探讨节律基因在慢性粒细胞白血病(CML)中的表达情况,及其与bcr/abl融合基因的相关性,为临床治疗提供新的分子靶点。方法收集106例CML患者慢性期、加速期、急变期的骨髓标本和50例健康体检人员的外周血为对照,采用RT-PCR法检测per1、per2、bcr/abl基因表达情况。结果 per1、per2、bcr/abl基因在慢性期、加速期和急变期存在不同水平的变化,并且per1、per2表达水平与bcr/abl基因存在负相关。结论节律基因作为肿瘤抑制因子在CML中是低表达的,在信号转导通路中是否与bcr/abl基因相互作用还需深入研究。Objective To investigate the expression level of circadian gene in the chronic myeloid leukemia and the relationship of bcr/ahl fusion gene,and provide the new molecular targets for the clinical treatment. Methods 106 cases of CML patients in chronic phase, accelerated phase, blast phase of bone marrow samples and 50 cases of normal peripheral blood were collected, the expression of perl,per2 and bcr/abl gene were detected by RT PCR. Results perl,per2 and bcr/abl gene are different levels in chronic phase,accelerated phase and blast phase, and perl, per2 gene are negative correlation with bcr/abl gene. Conclusion circadian gene is low in the expression of CML as a tumor suppressor, further research will need to confirm whether the bcr/abl gene interact with circadian gene in gignal transduction pathway.
