摘 要:目的探讨一种高效、简便和稳定的羊水细胞染色体制备方法,提高产前诊断的准确性和成功率。方法选择2002年9月3日到2009年7月9日孕周为16.0~29的产前筛查为高风险的孕妇2689位,抽取羊水进行细胞培养,行羊水细胞染色体核型分析。结果 2689例羊水细胞培养成功率为99.9%,核型分析的成功率为99.8%,发现异常核型74例,异常核型率为2.8%,其中18-三体10例,占0.37%,21-三体19例,占0.71%。结论本方法具有高效、简便、染色体带纹清晰等优点,在产前诊断中具有结果准确可靠和实用价值。Objective:To investigate a high effective,simple and steady amniotic fluid cells chromosomal preparation method,and to improve veracity and achievement ratio of prenatal diagnose.Methods:We cultured amniotic fluid cells and analysised them by chromosome karyotypes from 2689 prenatal-screening high risk women with 16.0~29 pregnant weeks since September 3,2002 to July 9,2009.Results:We found that the succeed proportion of 2689 amniotic fluid cells cultivate is 99.9%,the succeed proportion of karyotypes analysis is 99.8%,74 abnormal chromosomal karyotypes were detected,the abnormal tate was 2.8%,including 10 trisomy 18(0.37%),19 trisomy 21(0.71%).Conclusions:The method possessed many advantages such as high effect,simpleness and distinct karyotypings,and it has veracity and practicality in prenatal diagnose.
