摘　　要：目的:探讨环氧化酶-2(COX-2)在上皮性卵巢肿瘤的表达及环氧化酶-2抑制剂塞来昔布(celecoxib)在体外对人卵巢癌A2780细胞株增殖的影响。方法:①采用免疫组化SP法检测COX-2在65例上皮性卵巢癌、18例交界性与18例良性上皮性卵巢肿瘤及9例正常卵巢组织中的表达;②采用四甲基偶氮唑蓝(MTT)法观察塞来昔布在体外对A2780细胞增殖的影响;③流式细胞仪(FCM)测定塞来昔布在体外对A2780细胞周期的影响。结果:①COX-2在上皮性卵巢癌组(73.80%)及交界性卵巢肿瘤组的表达(66.70%)显著高于良性卵巢肿瘤组(16.70%)及正常卵巢组(11.10%),均P<0.05;②MTT显示,塞来昔布在体外以时间-剂量依赖方式抑制A2780增殖(P<0.05);③塞来昔布作用72 h后A2780细胞周期呈剂量依赖方式出现G0/G1期阻滞(P<0.05)。结论:①COX-2在交界性及恶性上皮性卵巢肿瘤中的表达明显增强,COX-2可能在交界性及恶性上皮性卵巢肿瘤的发生、发展中起一定作用;②体外特异性COX-2抑制剂(塞来昔布)能够抑制卵巢癌A2780细胞株增殖,有可能成为卵巢癌化疗的有效药物。Objective：To investigate the expression of cyclooxygenase-2（COX-2） in epithelial ovarian carcinoma tissues and to investigate the effects of Celecoxib on the cell proliferation of the human ovarian carcinoma cell line A2780. Methods：①The immunohistochemical was used to determine the expression of COX-2 in 65 primary ovarian carcinoma,18 low magligant potential tumors,18 cystadenomas and 9 normal ovaries.②Detect the effects of Celecoxib on the growth of ovarian carcinoma cell lines by MTT methods.③Detect the effects of Celecoxib on F1owCytometry（FCM）,cell cycle prohibition of ovarian carcinoma cells. Results：①The expression of COX-2 in epithelial ovarian cancer tissue and in borderline tumor tissue was remarkably higher than that in benign epithelial ovarian tumor tissues and normal ovarian tissue（P0.05）.②Proliferation of A2780 cell line was significantly inhibited by Celecoxib in some time and dosage dependent manner（P0.05）.③ FCM showed A2780 cell line was significantly inhibited by Celecoxib in 72 hours in some dosage dependent manner,Celecoxib caused G0/G1 arrest of A2780 cell line（P0.05）. Conclusion：① The expression of COX-2 is higher in the tissue of malignant and borderline epithelial ovarian tumor.② In vitro,celecoxib can suppress the proliferation of human ovarian adenocarcinoma cell line A2780 and inhibit cell cycle of A2780.The COX-2 inhibitor may be a novel which is useful chemoprevention agents for human ovarian cancer.
