实时逆转录荧光定量聚合酶链反应检测外周血GPC3mRNA在原发性肝癌患者中的表达及意义

摘　　要：目的探讨实时逆转录荧光定量聚合酶链反应(RealtimeQRT-PCR)检检测外周血磷脂酰肌醇蛋白聚糖3 mRNA(GPC3 mRNA)在原发性肝癌(HCC)患者中的表达意义。方法采用Realtime QRT-PCR技术分别检测37例HCC、21例转移性肝癌、22例肝硬化患者和35例体检健康志愿者外周血中GPC3 mRNA的水平。结果 HCC患者外周血GPC3mRNA的阳性率为91.8%(34/37),表达水平为(1.5±0.9)×106copies/ml;转移性肝癌外周血GPC3 mRNA的阳性率为4.7%(1/21),表达水平为5.2×103copies/ml;肝硬化患者GPC3 mRNA的阳性率为4.5%(1/22),表达水平为2.6×103 copies/ml;体检健康志愿者外周血GPC3 mRNA无阳性表达。HCC患者手术前后血液中GPC3 mRNA的阳性率差异无统计学意义(P<0.05),但两者表达水平差异有统计学意义(P<0.05)。结论 Realtime QRT-PCR检测外周血GPC3 mRNA是诊断早期HCC血微转移可行的实验方法。Objective To explore expression of GPC3 mRNA in perpheral blood detected by Realtime quantitative（Realtime QRT-PCR） among patients with hepatocellular carcinoma（HCC） and its significance.Methods GPC3mRNA in perpheral blood were detected by Realtime RT-PCR in 37 patients with HCC,21 with metastatic liver cancer,22 with cirrhosis and 35 health volunteers.Results GPC3 mRNA positive rate in HCC patients was 91.8%（34/37）,and the expression level was（1.5€？.9）€？06 copies/ml;GPC3 mRNA positive rate in metatastic liver cancer was 4.7%（1/21）,and the level was 5.2×103copies/ml;GPC3 mRNA positive rate in cirrhosis patients was 4.5%（1/22）,and the level was 2.6×103 copies/ml;GPC3 mRNA in health volunteers was negative.Conclusions ealtime QRT-PCR is a sensitive and reliable method for detecting circulating HCC cells.
