摘 要:目的:探讨中医活血健脾补肾法对骨髓间充质干细胞(MSC)移植在结肠炎大鼠肠道定植的影响。方法:雌性SD大鼠以5%2、4、6,三硝基苯磺酸(TNBs)诱导形成结肠炎模型,造模后随机分为模型对照组、MSC移植组、活血组、健脾组、补肾组,第2天将体外分离培养的雄性SD大鼠MSC经尾静脉注射到各组雌性大鼠体内。疗程结束后取各组大鼠结肠组织,以Y染色体的性别决定区段sry作为标志,以确定MSC的肠道定位情况;免疫组化法检测其肠道干细胞标记物Musashi-1的表达。结果:经PCR扩增后,凝胶电泳可以检测到结肠组织雄性性别特异性基因区段sry,MSC移植组阳性率为20%,活血组阳性率为35%,健脾和补肾组阳性率为25%。免疫组化检测到MSC移植组及中药治疗各组大鼠结肠黏膜组织Musashi-1均有阳性表达,其中活血组与MSC移植组比较差异有显著性意义。结论:活血法代表方药桃红四物汤较明显促进MSC在结肠炎大鼠肠道定植。Objective To explore the effect of promoting blood flow, invigorating spleen or invigorating kidney recipe on localization and differentiation of bone marrow mesenchymal stem cells (MSCs) in the colon of ulcerative colitis (UC) after transplantation. Methods Female UC model SD rats were induced by 2,4,6- trinitrobenzenesulfonic acid acid (TNBs) with the concentration of five percent, then were divided randomly into the model group, the MSCs transplantation group, promoting blood flow group, invigorating spleen group andinvigorating kidney group. The next day, MSCs of male SD rats cultured and purified in vitro were transplanted into the female UC model rats through caudal vein. After treatment, the rats colon tissue were taken Io identify the intestinal localization of MSCs by polymerase chain reaction (PCR), using sry section, the sex-determining region in Y chromosome, as a symbol. The expression of Musashi-1, the intestiml stem cell marker, was detected by immunohistochemical method. Results The expression of male sex-specific gene segments sry in Colon tissue could be detected by gel electrophoresis after PCR amplification. The positive rate of MSCs transplantation group, promoting blood flow group, invigorating spleen group and invigorating kidney group were 20%, 35%, 25% and 25% respectively. The expression of Musashi-1 in MSCs transplantation group and those TCM treatment groups was positive. Comparing with MSCs transplantation group, the expression of Musashi-1 in promoting blood flow group is significantly increased (P〈0.05). Conclusion Promoting blood flow recipe could promote the localization and differentiation of MSCs in the colon of colitis rat.
