摘  要:目的:分离鉴定葛根糖基转移酶蛋白肽段序列。方法:从野葛根部提取糖基转移酶,分离后对符合糖基转移酶分子量大小的5条蛋白条带进行酶解,经过高效液相色谱分离纯化后,电喷离子化质谱测定获得蛋白。结果:共获得440蛋白,发现有明确功能的蛋白有325个;具有催化活性的蛋白质有247个。根据报道的糖基转移酶分子量和等电点分布的特点,筛选出6个可能为糖基转移酶的蛋白。结论:初步推测葛根糖基转移酶的蛋白肽段序列,为其在葛根素生物合成中的作用研究奠定基础。Objective:Separation and identification of peptides sequence of glucosyltransferase in Pueraria lobata(Willd.) Ohwi.Method:Extract of glucosyltransferase from the roots of Pueraria lobata(Willd.)Ohwi was separated by SDS-PAGE,and 5 protein bands in accordance with molecular mass of glucosyltransferase were digested by enzymes.After separation by HPLC,the enzymatic product were sequenced by LC-ESI-MS/MS.Result:Totally,440 proteins were obtained.325 proteins were annotated,247 proteins possess catalytic activity.According to the characteristics of the molecular mass and isoelectric point distribution of glucosyltransferase reported,6 putative glucosyltransferases were screened.Conclusion:Preliminary speculating peptides sequence of glucosyltransferase for the study on puerarin biosynthesis.
