葛根糖基转移酶蛋白肽段序列的分离与鉴定

摘　　要：目的:分离鉴定葛根糖基转移酶蛋白肽段序列。方法:从野葛根部提取糖基转移酶,分离后对符合糖基转移酶分子量大小的5条蛋白条带进行酶解,经过高效液相色谱分离纯化后,电喷离子化质谱测定获得蛋白。结果:共获得440蛋白,发现有明确功能的蛋白有325个;具有催化活性的蛋白质有247个。根据报道的糖基转移酶分子量和等电点分布的特点,筛选出6个可能为糖基转移酶的蛋白。结论:初步推测葛根糖基转移酶的蛋白肽段序列,为其在葛根素生物合成中的作用研究奠定基础。Objective：Separation and identification of peptides sequence of glucosyltransferase in Pueraria lobata（Willd.） Ohwi.Method：Extract of glucosyltransferase from the roots of Pueraria lobata（Willd.）Ohwi was separated by SDS-PAGE,and 5 protein bands in accordance with molecular mass of glucosyltransferase were digested by enzymes.After separation by HPLC,the enzymatic product were sequenced by LC-ESI-MS/MS.Result：Totally,440 proteins were obtained.325 proteins were annotated,247 proteins possess catalytic activity.According to the characteristics of the molecular mass and isoelectric point distribution of glucosyltransferase reported,6 putative glucosyltransferases were screened.Conclusion：Preliminary speculating peptides sequence of glucosyltransferase for the study on puerarin biosynthesis.
