摘  要:目的:研究内毒素血症发生时多种脂多糖(LPS)相关蛋白在LPS转运及活化中的调节作用。方法:培养佛波醇酯(PMA)诱导分化的THP-1细胞。选取LPS、脂多糖结合蛋白(LBP)、可溶性CD14(sCD14)、杀菌/渗透增强蛋白(BPI)、高密度脂蛋白(HDL)5因素2水平采用L16(215)正交设计。配制实验体系,培养4 h后,留取上清液,发光法检测TNF-α分泌量。结果:混合体系中,BPI和HDL存在负向效应,LBP、LPS和sCD14存在正向调节效应。TNF-α分泌量与sCD14、BPI的单独主效应有关(P<0.05),并与LBP和sCD14、LBP和HDL、LPS和LBP的交互作用有关(P<0.05)。结论:在体内多种相关蛋白同时存在时,LPS对单核巨噬细胞的活化与sCD14、BPI的单独效应有关,并与LBP和sCD14、LBP和HDL、LPS和LBP交互作用有关。Objective: To study a variety of LPS related proteins regulatory effect on LPS transfer and activation in endotoxemia.Methods: PMA-differentiated THP-1 cells were cultured and five reagents of LPS,LBP,sCD14,BPI,HDL with two levels were selected through L16(215)orthogo-nal design table.A mixed reagents system was prepared and added into the cell,cultured for 4 hours,aspirated the supernatant to detect TNF-α.Results: In the mixed system,BPI and HDL had negative effect,while LBP,LPS,and sCD14 had positive effects.TNF-α secretion by THP-1 cell was related with main effect of sCD14 and BPI(P 0.05),as well as interaction between LBP and sCD14,LBP and HDL,LPS and LBP(P 0.05).Conclusion: In the body that LPS-related proteins simultaneously exist,activation of mononuclear macrophage cell induced by LPS is related with main effect of sCD14 and BPI,as well as interaction between LBP and sCD14,LBP and HDL,LPS and LBP.
