摘 要:目的:探讨卵巢癌缺氧诱导因子HIF-1α、基质金属蛋白酶MMP-2的表达及其与血管生成拟态(VM)的关系。方法:收集本院2000年1月~2009年5月经手术切除的卵巢肿瘤组织石蜡标本81例,免疫组化检测HIF-1α、MMP-2蛋白表达,采用CD34和PAS双重染色的方法检测卵巢癌组织中存在的VM,讨论HIF-1α、MMP-2表达与VM的关系。结果:卵巢癌HIF-1α表达与临床分期(r=0.392,P=0.020)、分级(r=0.358,P=0.035)呈正相关,MMP-2表达与分级(r=0.409,P=0.015)呈正相关。卵巢癌不同分级组的VM形成率有显著差异(P=0.018),低分化的卵巢癌VM阳性率高于高分化组。VM阳性组和阴性组的HIF-1α表达有统计学差异(P=0.003),VM阳性组和阴性组的MMP-2表达有统计学差异(P=0.004)。结论:低分化卵巢癌组织更易形成VM,HIF-1α、MMP-2表达与卵巢癌分级及VM密切相关。Objective:To investigate the relationship between expression of hypoxia induced factor-1α(HIF-1α),matrix metalloproteinase-2(MMP-2) and vasculogenic mimicry(VM).Methods:81 paraffin-embedded sections of ovarian tumor were studied.Expression of HIF-1α and MMP-2 were detected by immunohistochemical staining.The presence of VM was observed by histochemical double staining of CD34 and PAS.Relationship between expression of HIF-1α,MMP-2 and VM were discussed.Results:Positive expressions of HIF-1α and MMP-2 protein were detected in ovarian cancer.Vasculogenic mimicry,when PAS was positive but CD34 was negative,appeared in some cases of ovarian cancer.Positive correlation was observed between tumor grade and VM(P=0.018).Positive relationship was found between expression of HIF-1α and clinical stage(r=0.392,P=0.020),grade(r=0.358,P=0.035).Positive relationship was also found between expression of MMP-2 and grade(r=0.409,P=0.015).Expression of HIF-1α(P=0.003) and MMP-2(P=0.004) in ovarian cancer with VM were increased significantly than those without VM.Conclusion:Vasculogenic mimicry exists in ovarian cancer,especially in the lower grade.Expression of HIF-1α and MMP-2 are associated with tumor grade and formation of vasculogenic mimicry.
