雷公藤内酯醇对大鼠大脑皮质内注射β-淀粉样蛋白后补体C1q和C3表达的影响

摘　　要：目的探讨雷公藤内酯醇对大鼠大脑皮质内注射β-淀粉样蛋白(Aβ)后补体C1q和C3表达的影响。方法 30只SD雄性大鼠随机等分成对照组、Aβ组、用药组。Aβ组大鼠给予双侧大脑皮质各一次性注射凝聚态Aβ1-40,对照组大鼠大脑皮质注射等量生理盐水,用药组大鼠在大脑皮质注射凝聚态Aβ1-40后腹腔注射雷公藤内酯醇,免疫组织化学染色和RT-PCR方法检测各组大鼠大脑皮质C1q和C3蛋白和mRNA的表达。结果免疫组织化学染色显示,Aβ组大鼠大脑皮质C1q和C3免疫反应阳性细胞数和平均光密度明显高于对照组(P<0.01);用药组大鼠大脑皮质C3免疫反应阳性细胞数较Aβ组明显减少(P<0.05),用药组大鼠大脑皮质C1q和C3免疫反应阳性细胞平均光密度较Aβ组明显减弱(P<0.01)。RT-PCR结果显示,Aβ组大鼠大脑皮质C1q和C3 mRNA表达量明显高于对照组(P<0.01);用药组大鼠大脑皮质C1q和C3 mRNA表达量明显低于Aβ组(P<0.05,P<0.01)。结论雷公藤内酯醇对大鼠大脑皮质内注射Aβ后补体C1q和C3的表达有抑制作用。Objective To explore the effects of triptolide on the expressions of C1q and C3 after microinjection of beta-amyloid protein（Aβ） into cerebral cortex in rats.Methods Thirty male SD rats were equally divided into control,Aβ and triptolide-treated groups.The Aβ group was made with bilateral microinjection of aggregated Aβ1-40 into cerebral cortex in rats and control group rats were injected with normal saline in the same way.The triptolide-treated group rats were administered triptolide intraperitoneally after microinjection of aggregated Aβ1-40 into cerebral cortex.The protein and mRNA expressions of C1q and C3 of cerebral cortex in each group were assayed by immunohistochemical staining and reverse transcription polymerase chain reaction（RT-PCR）.Results The cell number and average optical densities of C1q and C3 positive staining in Aβ group were higher than those in control group（P0.01）,cell number of C3 positive staining in triptolide-treated group was lower than that in Aβ group（P0.05）,the average optical densities of C1q and C3 positive staining in triptolide-treated group were lower than those in Aβ group（P0.01）.In the experiment of RT-PCR,mRNA expressions of C1q and C3 in Aβ group were higher than those in control group（P0.01）.The mRNA expressions of C1q and C3 in triptolide-treated group were lower than those in Aβ group（P0.01）.Conclusions Triptolide can inhibit the expressions of C1q and C3 after microinjection of Aβprotein into cerebral cortex in rats.
