β_1-整合素与膜受体PTCH在大肠癌中的表达及其相互关系研究

摘　　要：目的:检测β1-整合素(β1-integrin)及hedgehog(Hh)信号通路中膜受体蛋白patched(PTCH)在不同分化程度结直肠腺癌中的表达,探讨其表达与肿瘤生物行为学间的关系并分析两者间的相关性。方法:应用免疫组化SP法检测组织芯片69例结直肠腺癌中β1-integrin与PTCH的表达情况,并结合临床病理资料进行分析。结果:β1-integrin、PTCH的阳性表达率分别为59.42%(41/69)和63.77%(45/69),其中β1-integrin的表达与肿瘤的Dukes分期、浸润深度、淋巴结转移呈正相关(P<0.05),并且两者的阳性表达呈明显的正相关性(P<0.05)。结论:β1-integrin与结直肠腺癌的发生及浸润转移密切相关,并可能通过增加Hh通路上游蛋白PTCH水平的表达而激活该信号通路。Objective： To investigate the expression of β1-integrin and the transmembrane protein receptor patched（PTCH） of hedgehog（Hh） signaling pathway in colorectal adenocarcinoma,and to explore the interaction between PTCH and β1-integrin as well as their correlation to the biological behavior of carcinoma.Methods： Immunohistochemical method of SP was used to examinate the expression of β1-integrin and PTCH in 69 cases of colorectal adenocarcinoma.Results： The positive expression rates of β1-integrin and PTCH were 59.42%（41/69） and 63.77%（45/69） respectively.Significant differences in the expression of β1-integrin were found in different depth of invasion,lymph node metastasis,and Dukes staging（all P0.05）.And positive relationship was also found between PTCH and β1-integrin expression（P0.05）.Conclusion： The expression of β1-integrin is significantly correlated to the development,invasiveness,and metastasis of colorectal adenocarcinoma,and it might activate the Hh signaling pathway through increasing the expression of PTCH.
