摘  要:目的探讨姜黄素对原代培养的兔结膜上皮细胞增殖的影响,为预防翼状胬肉术后复发探讨新的策略。方法制备原代兔结膜上皮细胞,以不同浓度的姜黄素(0、10、20、40、60μmol/L)作用于体外培养的第3~5代结膜上皮细胞,分别于处理后24、48、72h后采用MTT法检测细胞增殖情况。此外,采用上述浓度姜黄素处理结膜上皮细胞24h后,采用流式细胞仪检测细胞周期变化。结果 20μmol/L以上浓度的姜黄素可以显著抑制兔结膜上皮细胞增殖,且随浓度增加效果增强,随着时间增加抑制率显著增加(P<0.05)。此外,姜黄素作用24h后的细胞周期变化结果显示,G0/G1期细胞数量增加,细胞增殖受到显著抑制(P<0.05)。结论姜黄素能够抑制体外培养的原代兔结膜上皮增殖,姜黄素有可能作为抑制翼状胬肉术后复发的药物。Objective To investigate the inhibitory effect of curcumin on primary rabbit conjunctival epithelium cells and to explore a new way to prevent postoperative recurrence of pterygium.Methods The third to fifth passage of primary rabbit conjunctival epithelium cells were prepared and cultivated with curcumin of different concentrations(0,10,20,40 and 60 umol/L).Inhibitory effect of curcumin on primary rabbit conjunctival epithelium cells was measured by MTT method after 24,48 and 72 hours,respectively.Flow cytometry was used to measure cell cycle after cultivation with curcumin of different concentrations for 24 hours.Results Curcumin with concentration greater than 20 umol/L could significantly inhibit the proliferation of rabbit conjunctival epithelium cells and the effect was more prominent as time went on(P0.05).After cultivation with curcumin for 24 hours,more cells tended to stay at G0/G1 cell cycle and the proliferation was significantly inhibited(P0.05).Conclusion Curcumin inhibited proliferation of primary rabbit conjunctival epithelium cells through inhibition of cell cycle and might be a promising drug used for postoperative pterygium recurrence.
