摘  要:目的调查精神分裂症患儿神经营养素-3基因(NT-3)甘氨酸(Gly)/谷氨酸(Glu)多态性与脑室扩大及微小躯体异常(MPAs)之间的关系。方法采用聚合酶链反应-限制性片段长度多态性方法分析218例精神分裂症患儿和210例健康儿童NT-3基因Gly/Glu多态性;采用CT测量患儿的脑室参数(计算哈氏值、三脑室宽度、脑室指数、侧脑室体部指数、侧脑室宽度指数、前角指数表示),采用waldrop编制的躯体异常量表(WS)修改版评定MPAs发生情况;采用方差分析、协方差分析、t检验比较NT-3不同基因型(Gly/Gly、Gly/Glu、Glu/Glu)患儿之间脑室值的差异,采用非参数检验及t检验比较不同基因型患儿之间的WS评分差异,脑室值与WS评分的相关关系采用Spearman相关分析。结果 1.研究组NT-3基因Gly/Gly频率高于对照组,差异有统计学意义(75.2%vs64.3%,χ2=6.86,P<0.05),等位基因Gly频率高于对照组,差异有统计学意义(86.2%vs81.0%,χ2=4.37,P<0.05)。2.不同基因型患儿之间各脑室值比较差异均无统计学意义(Pa>0.05)。3.Glu/Glu基因型患儿头部WS评分显著高于Gly/Gly和Gly/Glu型患儿(Z=2.237,2.728,Pa<0.05)。4.WS总分与哈氏值及三脑室宽度均呈正相关(r=0.26、0.29,Pa<0.01),与前角指数呈负相关(r=-0.178,P<0.05)。结论精神分裂症患儿NT-3基因Gly/Glu多态性与微小躯体异常有关,与脑室扩大之间无显著关系。Objective To investigate the relationships between glycine/glutamic acid(Gly/Glu) polymorphisms of neurotrophin 3(NT-3) gene and entricular enlargement as well as minor physical anomalies in childhood schizophrenia.MethodsThe Gly/Glu polymorphisms of NT-3 gene were genotyped in 218 childhood schizophrenia and 210 normal subjects with polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP) method.Morphological abnormalities of brain were evaluated with ventricular-brain value by using cerebral computed tomography(CT) examination.Ventricular-brain value consisted of Huckman′s value,extend of the third ventricle,index of ventricle,index of lateral ventricle body,index of extend of lateral ventricle and index of anterior angle of ventricle.Their minor physical anomalies were examined on the Waldrop Scale(WS).The difference on ventricular-brain value among different genotype of NT-3 gene was compared with analysis of variance and analysis of covariance and t test,the difference on WS score among different genotype of NT-3 gene was compared with nonparametric test and t test.The relationship between WS score and ventricular enlargement was analyzed with Spearman correlation analysis.Results1.The frequency of the Gly/Gly genotype and Gly allele were significantly higher in childhood schizophrenia(75.2%,86.2%,respectively) compared with the controls(64.3%,81.0%,respectively,χ2=6.86,4.37,Pa0.05).2.There was no significant difference among patients with different genotype groups in values of every ventricular-brain(Pa0.05).3.The patients with Glu/Glu genotype had significantly higher value on score of WS for head than Gly/Gly and Gly/Glu genotypes(Z=2.237,2.728,Pa0.05).4.Total score of WS was significantly positive correlated with Huckman′s value and extend of the third ventricle(r=0.26,0.29,Pa0.01) and negatively correlated with index of anterior angle(r=-0.178,P0.05).ConclusionsThese results indicate that the Gly/Glu polymorphism of the NT-3 g
