摘  要:目的:建立同时测定通脉颗粒中葛根素和阿魏酸含量的快速分析方法。方法:采用HPLC-UV法,选择KinetexC18柱(3 mm×50 mm,2.6μm)为分析柱,以10%乙腈-1%醋酸(54∶46)为流动相,流速0.6 mL.min-1,313 nm检测波长下同时检测葛根素和阿魏酸。结果:在此分析条件下,葛根素和阿魏酸的保留时间分别为5.7,7.6 min,与阴性样品对照具有特异性,一次分析可在10 min内完成。葛根素线性范围在10~100 mg.L-1,r=0.999 7;阿魏酸线性范围在0.05~0.5 mg.L-1,r=0.999 3;葛根素和阿魏酸的平均加样回收率分别达到(98.1±1.4)%,(103.7±1.7)%。结论:该方法快速、灵敏、准确、精密,可以为通脉颗粒剂的质量控制提供快速的含量测定方法。Objective:To achieve rapid quantitation of active components in Tongmai granule.Method: By choosing Kinetex C18(3 mm × 50 mm,2.6 μm) as analysis column,an HPLC-UV method was established to detect puerarin and ferulic acid in Tongmai Granule under detection wavelength of 313 nm.The mobile phrase was optimized as 10% acetonitrile-1% acetic acid(54∶ 46) at flow rate of 0.6 mL.min-1.Result: Under this analytic condition,the quantitation could be accomplished within 10 minutes profiling good separation and specificity of puerarin and ferulic acid which presented retention time at 5.7,7.6 min.The average recovery was(98.1 ± 1.40)%,(103.7 ± 1.68) % for puerarin and ferulic acid respectively.Calibration curves ranged from 10 to 100 mg.L-1for puerarin and from 0.05 to 0.5 mg.L-1 for ferulic acid with correlation coefficients of 0.999 7 and 0.999 3.Conclusion:The method is rapid,sensitive,accurate and precise.It could be used to provide a rapid determination in quality control of Tongmai granule.
