电针治疗对大鼠局灶性脑缺血ERK磷酸化的调节作用

摘　　要：目的探讨电针对局灶性脑缺血大鼠顶叶皮质细胞外信号调节激酶(ERK)蛋白磷酸化的影响。方法采用线栓法制作大鼠大脑中动脉闭塞(MCAO)模型,将90只雄性SD大鼠随机分为3组,即对照组、实验组、治疗组;治疗组以韩氏穴位神经刺激仪电针"百会""水沟"穴,每日1次,留针30min,治疗15d;治疗结束后用神经行为学评分标准评定大鼠的神经功能;应用实时定量PCR、Western Blotting和图像分析方法检测缺血侧顶叶皮质ERK1和ERK2的表达水平。结果经电针治疗后脑缺血大鼠损伤的神经行为学评分得到不同程度的改善。ERK1/2表达在大鼠局灶性缺血脑组织中显著升高。结论电针对大鼠局灶性脑缺血的ERK磷酸化起着重要的调节作用,并对缺血性脑损伤有一定保护作用。Objective To explore the effect of electric acupuncture on phosphorylation effect of extracellular signal regulated kinase（ERK）in focal cerebral ischemia.Methods Ninety male SD rats were randomized into control,model group,and treatment group.Focal cerebral ischemia model was induced by occlusion of the middle cerebral artery using the intraluminal suture method.＇Baihui＇（GV20） and ＇Shuigou＇（GV26） were punctured and stimulated by twirled the acupuncture needle with Han＇s acupoint nerve stimulator continuously to maintain the needle for 30 min.The treatment was conducted once daily and continuously for 15 days.The score of neuroethology measured after the cerebral ischemia model and after the treatment.The expressions of ERK in the ipsilateral parietal cortex of rats were detected with Real-time quantitative PCR and Western Blotting.The positive products of ERK1 and ERK2 were analyzed by image analysis system.Results The neuroethology score changed significantly before and after treatment in treatment group（P0.01）.After treatment with electric acupuncture,the quantities of both ERK1 and ERK2 were found to be higher than that of ischemia model groups（P0.05）.Conclusions The results indicate that electric acupuncture can up-regulate the expression of ERK1 and ERK2 in the ipsilateral parietal cortex after focal cerebral ischemia and ERK may be involved in the protective mechanism of electric acupuncture to ischemic neurons.
