培哚普利及高糖对RF/6A细胞SOCS3表达的影响

摘  要:目的探讨培哚普利及高浓度葡萄糖对猴视网膜脉络膜内皮细胞(RF/6A)中细胞因子信号抑制因子3(suppressor of cytokine signaling3,SOCS3)表达的影响及机制。方法 RF/6A细胞分别用高浓度葡萄糖(25mmol.L-1)孵育0h、2h、4h、8h、24h、48h;用高糖(25mmol.L-1)、培哚普利(10μmol.L-1)、Janus激酶(janus kinase2,JAK2)抑制剂AG490(10μmol.L-1)、活性氧清除剂N-乙酰半胱氨酸(NAC,10mmol.L-1)分别干预24h后,采用Western blotting及RT-PCR检测细胞中SOCS3蛋白及mRNA的表达。结果 正常培养条件下,RF/6A细胞中SOCS3蛋白低水平表达,高糖刺激2h、4h、8h、24h均较0h明显升高(P<0.001),高糖刺激48h时下降到正常水平,与0h相比差异无统计学意义(P>0.05)。AG490、NAC均可抑制高糖作用后SOCS3蛋白和mRNA的表达升高,与高糖组相比差异均有统计学意义(均为P<0.05)。培哚普利则可显著上调RF/6A细胞中SOCS3蛋白及mRNA表达,与高糖组相比差异均有显著统计学意义(均为P<0.001)。结论 高糖及培哚普利可显著上调视网膜脉络膜血管内皮细胞中SOCS3表达;培哚普利的这一作用可能与其对ROS/JAK2/STAT3通路的抑制有关。Objective To determine the effects and mechanisms of high glucose and perindopril on suppressor of cytokine signaling 3(SOCS3) expression in cultured rhesus monkey choroid-retinal(RF/6A) cells.Methods RF/6A cells were incubated in high glucose(HG,25 mmol·L-1) for 0 hour,2 hours,4 hours,8 hours,24 hours,48 hours,respectively.After exposed to perindopril(10 μmol·L-1),Janus kinase 2(JAK2) inhibitor AG490(10 μmol·L-1)and reactive oxygen species(ROS) scavenger N-acetylcysteine(NAC,10 mmol·L-1) for 24 hours,respectively,RT-PCR and Western blotting method were used to assess the mRNA and protein expression of SOCS3 in RF/6A cells of each group.Results SOCS3 protein expression was very low in the normal RF/6A cells,it increased after induced by HG for 2 hours,4 hours,8 hours,and 24 hours,which was statistically significant compared with 0 hour group(All P0.001).But for 48 hours,there showed no significant difference compared with 0 hour group(P0.05).AG490 and NAC were able to decrease the expression of SOCS3 protein and mRNA in the cells that exposed to HG,and there were statistical differences compared with HG group(All P0.05).Perindopril increased the SOCS3 expression at both protein and mRNA level in RF/6A cells obviously,there were significant differences compared with HG group(All P0.001).Conclusion HG and perindopril can obviously increase the expression of SOCS3 in RF/6A cells.This mechanism of perindopril may be attributed to its suppression of ROS/JAK2/STAT3 pathway.
